基于Imaris 3D图形渲染方法对神经元形态量化分析

摘要 目的利用Imaris软件对在体稀疏标记神经元及体外培养原代神经元所收集的实验数据进行多种统计量化分析。方法利用Imaris软件的Filament模块、Spots模块、Surface模块,对神经科学领域中涉及神经元的荧光采集图像进行3D渲染并分析数据,包括质量检查、去除背景、神经元重构、胞体测量、突触棘分型统计以及体外培养神经元的量化统计。结果基于Imaris的在体神经元的量化统计结果显示,各级神经元平均体积存在显著差异,长度和面积差异无统计学意义;不同分型突触棘表面积差异存在统计学意义。结论Imaris 3D图形渲染方法可有效应用于神经元及树突棘形态重构、神经元胞体测量等量化分析中,从而为神经生物学的精准量化分析提供有力手段。 Objective To study how Imaris software can be used for a variety of statistical quantitative analysis on the experimental data collected from sparsely labeled neurons in vivo and cultured primary neurons in vitro.Methods The Filament module,Spots module and Surface module of Imaris software were used to render and analyze the fluorescence images of neurons in 3 D,involving quality inspection,background removal,neuron remodeling,cell body measurement,synaptic spine typing statistics and quantitative statistics of neurons cultured in vitro.Results The quantitative statistical results of neurons in vivo based on Imaris showed that there were significant differences in the average volume of neurons at each level,but there was no significant difference in the length and area of neurons,and there were statistically significant differences in the surface area of synaptic spines between different types.Conclusion Imaris 3 D graphics rendering method can be effectively applied to quantitative analysis of morphological reconstruction of neurons and dendritic spines and measurement of neuronal cell bodies,thus providing a powerful means for accurate quantitative analysis of neurobiology.
