2650 MHz射频辐射对C57BL/6N小鼠情绪及学习记忆的影响研究

摘要 目的探讨2650 MHz射频辐射对C57BL/6N小鼠情绪及学习记忆的影响。方法采用电磁混响室对20只小鼠进行射频电磁波暴露,暴露的比吸收率(SAR)为4 W/kg,4 h/d,连续暴露28 d。在暴露28 d后进行旷场实验和水迷宫实验,分别观察小鼠情绪行为和学习记忆能力;采用HE和TUNEL染色法观察小鼠海马组织神经元形态及凋亡变化;采用Western印迹检测海马组织脑源性神经营养因子(BDNF)的表达。结果在旷场实验中,与对照组相比,暴露组小鼠在总路程、中央区域路程和中央区域时间均有下降趋势,但无统计学差异(P>0.05);水迷宫结果显示,与对照组相比,暴露组小鼠的逃逸潜伏期显著增加;穿越原平台次数和目标平台象限停留时间百分比均显著降低(P<0.05)。HE染色显示,与对照组相比,暴露组小鼠海马齿状回神经元核固缩、深染明显。TUNEL染色显示,暴露组小鼠海马齿状回神经元凋亡明显。Western印迹结果显示,暴露组小鼠海马组织中BDNF表达显著降低(P<0.05)。结论2650 MHz射频辐射可导致小鼠学习记忆损伤,并与海马组织结构损伤、神经细胞凋亡以及BDNF表达降低相关。 Objective To investigate the effect of 2650 MHz radiofrequency exposure on the emotional behavior as well as learning and memory of C57 BL/6 N mice.Methods Twenty mice were kept exposed to radiofrequency electromagnetic fields in an electromagnetic reverberation chamber for 28 days,with a specific absorption rate(SAR)value of4 W/kg,4 hours per day.The open field test and Morris water maze test were performed after 28-day exposure to observe the changes in emotional behavior and learning and memory ability of mice respectively.HE staining and TUNEL staining were used to observe the morphological changes and apoptosis of hippocampal neurons in mice.Western blot was used to detect the expression of BDNF in the hippocampus.Results In the open field test,the total distance,the distance in the central area,and the time spent in the central area of the exposure group decreased compared with the control group,but there was no statistically significant difference(P>0.05).The water maze results showed that the escape latency of the mice in the exposure group was significantly increased compared with the control group,and the number of crossings of the original platform and the percentage of stay time in the quadrant of the target platform were significantly reduced(P<0.05).The results of HE staining suggested that,the normal shape of hippocampal neurons was seen in the control group,and the pyknosis and deep staining of hippocampal neurons in the exposure group were obvious.TUNEL staining results showed that the hippocampal dentate gyrus neurons in the exposed group had obvious apoptosis.Western blot showed that the expression of BDNF in the hippocampus of the exposed group was significantly reduced(P<0.05).Conclusion The2650 MHz radiofrequency exposure could cause serious damage to the learning and memory of C57 mice,which is related to hippocampal tissue structural damage,neuronal apoptosis and decreased expression of BDNF.
