生物毒素重组抗体的筛选及应用研究进展 被引量:1

Research progress in screening and application of biotoxin recombinant antibodies

摘要 来自于动植物、微生物和海洋生物的生物毒素对人类的生命及健康具有重要影响。自杂交瘤技术成功体外制备单克隆抗体以来,单抗逐渐成为科研、疾病诊断和治疗的重要工具和候选药物。同时,各种新的单抗筛选技术随之产生,其中噬菌体展示技术、酵母展示技术和单个B细胞技术在毒素重组抗体的筛选中得到了应用研究,而噬菌体展示技术以其经济、高效、简单易行、抗体特异性高等优点受到重点关注。该综述简要介绍了上述3种抗体筛选技术(特别是噬菌体展示技术)在生物毒素重组抗体筛选中的应用进展。 Biotoxins derived from animals,plants,microorganisms and marine organisms have an important effect on human life and health.Monoclonal antibodies have become pivotal tools in scientific research and drug candidates for disease diagnosis and treatment since they were prepared successfully by hybridoma technology in vitro.At the same time,a variety of new monoclonal antibody screening technologies have also emerged.For example,phage display,yeast display and single B cell technology have been applied in the screening of biotoxin recombinant antibodies.In particular,phage display has been increasingly used for screening biotoxin recombinant antibodies due to such advantages as low cost,high efficiency,simple operation and high specificity.Accordingly,this review introduces the three antibody screening technologies,especially phage display,and the progress in their applications in the screening of biotoxin recombinant antibodies.
