造血系统缺失Ronin基因降低胎肝细胞救治急性放射病的能力

摘要 目的研究造血系统特异敲除Ronin基因缺失后对胎肝细胞救治急性放射病能力的影响。方法利用基因组PCR和Western印迹鉴定Ronin的特异性敲除胚胎。8周龄CD45.1+C57BL/6J小鼠经60Co照射后,移植E14.5 d的野生型或敲除Ronin的小鼠(CD45.2+)胎肝细胞。移植后不同时间点检测受体小鼠存活率和体质量,并在移植后40 d分析外周血嵌合率。结果造血系统条件敲除Ronin基因后,胎肝细胞救治急性放射病的能力明显降低。结论Ronin是调节胎肝造血干细胞功能的关键因子,该研究为深入探讨Ronin基因在早期造血功能调控中的作用提供了实验模型。 Objective To evaluate the effect of Ronin ablation in the hematopoietic system on the ability of fetal liver cells to treat acute radiation injury. Methods Genomic PCR and Western blotting were used to identify Ronin-specific knockout embryos. Eight-week-old CD45.1+C57BL/6J mice with 60Co irradiation were injected with wild type or Ronin knockout cells from the E14.5 fetal liver(CD45.2+). Survival rate and body mass of the recipient mice were detected at different time points after transplantation,and 40 days after transplantation,the peripheral blood chimeric rate was analyzed.Results Ablation of Ronin gene in the hematopoietic system significantly reduced the ability of fetal liver HSCs to treat acute radiation injury.Conclusion Ronin is a key factor that regulates the function of hematopoietic stem cells of a fetal liver. This study provides an experimental model for further exploration of the role of Ronin gene in the regulation of early hematopoietic function.
