摘要
目的探讨高功率微波(HPM)辐射致大鼠及PC12细胞氧化损伤的效应,为HPM损伤模型的建立及防治药物的研究奠定基础。方法80只Wistar雄性大鼠随机分为4组,每组20只;PC12细胞随机分为4组。采用S波段微波源,以平均功率密度分别为0、10、30和100 mW/cm2辐照各组,大鼠辐照时间15 min,PC12细胞辐照时间6min。于辐射后6 h、1 d、3 d及7 d活杀大鼠并进行HE组织病理学染色和氧化应激指标比色法测量,包括超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-PX)、过氧化氢酶(CAT)和蛋白质羰基(PCO);细胞样品于24 h后进行活性氧(ROS)和Ca2+含量的流式细胞检测。结果10、30和100 mW/cm2HPM辐射后6 h^7 d可见大鼠海马神经元固缩深染,肝细胞不同程度变性肿胀,肝组织MDA和PCO含量明显增加,而GSH-PX和CAT活性明显下降;损伤效应程度随剂量升高而增大,随时间延长逐渐减轻;100 mW/cm2组辐射后24 h,PC12细胞ROS含量明显升高。结论一定条件的HPM辐射可致大鼠脑、肝及PC12细胞氧化应激损伤,且与剂量和时间有一定相关性。
Objective To investigate the effect of high power microwave(HPM)radiation on oxidative damage in rats and PC12 cells in order to contribute to research on the prevention and treatment of HPM.Methods Eighty Wistar rats were randomly divided into four groups with 20 in each.PC12 cells were also randomly divided into four groups.The Sband microwave source was used to irradiate each group at an average power density of 0,10,30 and 100 mW/cm2.The irradiation time of the rats was 15 min and that of PC12 cells was 6 min.After exposure,the rats were sacrificed at 6 h,1 d,3 d and 7 d for HE histopathology staining and colorimetric measurement of oxidative stress indexes,such as superoxide dismutase(SOD),malondialdehyde(MDA),glutathione peroxidase(GSH-PX),catalase(CAT)and protein carbonyl(PCO).Flow cytometry for PC12 cells was performed immediately after 24 hours to detect reactive oxygen species(ROS)and Ca2+content.Results After 10,30 and 100 mW/cm2 HPM radiation,the hippocampal neurons were condensed and deep-stained at 6 h-7 d,hepatocytes showed different degrees of degeneration and swelling,and the contents of MDA and PCO in liver tissue increased significantly,but the activities of GSH-PX and CAT decreased significantly.The degree of damage increased with the dose and gradually decreased with time.The ROS content of PC12 cells increased significantly 24 h after irradiation in the 100 mW/cm2 group.Conclusion Under certain experimental conditions,HPM radiation can cause oxidative stress damage in the rat brain,liver and PC12 cells.In addition,oxidative stress damage has some correlations with dose and time by HPM.
