摘要
哺乳动物受照射后骨髓组织的DNA合成受到明显抑制,其抑制程度与受照射剂量有密切关系,因此测定人体受照射部位的骨髓DNA合成能力对剂量诊断会有一定帮助。但此指标的临床应用受到取材量的限制。我们建立了用穿刺所得骨髓细胞离体进行~3H-TdR掺入并用同一标本同时测定其放射性及DNA含量的方法.
The estimation of DNA synthesis ability in bone marrow cells is of practical value in clinical investigation.But its application is restricted by the small amount of the sample available.A simple method for simultaneous estimation of the DNA content and radioactivity in same sample is described.The method consists of incorporating~3H-TdR in vitro,collecting the cells on glass fiber filter,counting the radioactivity by the liquid scintillating method,and quantitating the DNA content by diphenylamine method after treatment of the filter with hot perchloric acid,and then calculating the DNA specific radiactivity.An amount of 4×10~6 cells is sufficient for this assay.
