剥脱法估算药代动力学参数的FORTRAN—Ⅳ程序

摘要 许多药物在非静脉给药后的血药浓度曲线表现为三个明显时相:即血药浓度随时间而上升的吸收相;药物迅速向组织转运,使血药浓度迅速下降的分布(α)相;药物在体内达到拟分布平衡后,血药浓度按单指数式下降的清除(β)相。 A computer program,REGRE,has been developed for obtaining pharmacokinetic parameters from plasma drug concentration data after extravenous administration of drugs.This program was based on the standard residual method and written in FORTRAN.The program had following desirable characteristics:(a)Every possible combination of concentration data is calculated to estimate the coefficients and exponents of the polyexponential equation;(b)The goodness of fits is judged by the value of r and the sum of squares of the deviations between observed and estimated concentrations;The results are printed out automatically,including the regression equation,the pharmacokinetic parameters estimated;the values of observed and estimated drug concentrations,and the plasma drug concentration-time curve as well.A preliminary application of the program in studying pharmacokinetics of estrogen derivatives indicated that the program enables the rapid and accurate computer analysis of data and gives results better than those obtained by Laborious manual techniques.
