不同免疫活性细胞对照射小鼠移植骨髓的影响

摘要 同种造血细胞移植往往发生移植物排斥宿主(GVH)反应,后果也比较严重。例如,狗经1200伦照射后移植骨髓,DLA如果不相同,则绝大部分狗会发生严重的GVH病,即使DLA配型相同,仍会有部分狗发生此种并发病。狗照后输注同种外周血单个核白细胞,如事后不加免疫抑制措施. Adult male mice of Shanghai Strain were irradiated with 1000 rads ofγ-rays followed by transfusion of the same amount of cells from lymph node,thymus,spleen and bone marrow.The 30-day survival rate of the irradiated control group was about 5%.Mice treated with lymph node(LN)cells all died within 11 days after irradiation.The survival rate of mice treated with thymocytes was about 2—3%.but some mice in these groups died earlier than the irradiated controls.The mice of the spleen and bone marrow treated groups resulted in 42 and 57%survival respectively,but the addition of 5×10~6 or 2.5×10~6 LN cells to the bone marrow decreased the survival rate to 7%and 43%.Similar phenomenon was observed in mice transfused with bone marrow and spleen cells(2.5×10~6)simultaneously.Adult male mice of LACA strain were irradiated with 950 rads ofγ-rays followed by transfusion of same amount(1.5×10~6)of bone marrow or spleen cells.The survival rates were 77%and 45%respectively,but that of the former decreased to 52%when transfused in mixture with 9.5×1.0~6 spleen cells.LN-cells,which were irradiated with 4000 rads in vitro and transfused into lethally irradiated mice,aggravated the symptoms of irradiation injury leading to death at an earlier date than the control animals.Mice transfused with bone marrow cells together with irradiated LN-cells after irradiation showed a survival rate about 30%lower than that obtained from transfusion of bone marrow alone.
