应用微量固相放射免疫测验检测甲型肝炎抗原和抗体

APPLICATION OF MICRO-SOLID PHASE RADIOIMMUNOASSAY IN THE DETERMINATION OF HEPATITIS A ANTIGEN AND ANTIBODY

摘要 自70年代中期Hollinger及Purcell等先后报告可用(Micro-SPRIA检测甲型肝炎抗原(HAAg)和抗体(抗-HAV)后,这一作为甲型肝炎实验诊断的第三代技术,由于敏感、特异、操作简便和对HAAg纯度要求不高,故日益得到广泛应用。但在国内,近年来仅有少数单位应用该技术的初步报告。 A method of micro-solid phase radioimmunoassay(SPRIA)for detection of hepatitis A antigen and antibody was reported The optimum conditions for SPRIA were determined by comparison between different concentrations of anti-HAV IgG used to coat the solid phase plate and between different times taken for antigen-antibody incubation.The resul-ts of experimantal study showed that this method could specifically detect HAAg and anti-HAV,and its senstivity and reproducibility were high.In addition,the results ob rained from some of the examples(stools from a HA patient and sera from experimantally HAV infected monkeys)by this method were consistent with those detected by IEM and by IAHA or RIA(Abbott Laboratories,HAVAB).In 1980,the anti-HAV prevalence of 273 healthy soldiers in Peking was found by this method to be 77.3%.
