摘要
本实验应用单克隆抗胆碱乙酰化酶(ChAT)血清、卵白素—生物素复合体、AEC成色的免疫组织化学方法,对8只猫的骶尾髓进行了免疫组化染色观察。结果为:1.ChAT免疫反应阳性细胞:在L_(7)~Co_(1)节段前角各核团可见大量ChAT阳性神经元,它们可分为大、中、小3型。大、中型多极运动神经元的染色强度为淡至中等;小型神经元染色浓深,三者在核团内混杂存在。在骶髓中间带外侧核、中间带内侧核均可见梭形、圆形或椭圆形的副交感节前神经元,呈中等到深染;中介核内可见成色较淡的横向排列的长梭形神经元。在Ⅶ层内还可见成色深的中型多极神经元。在X层室周灰质中亦可见较深染的中、小型神经元。本文证明,后连合核区的腹侧部存在相当数量的小型阳性神经元,而在背侧部仅偶见于S2节段。后角偶见小型淡染的神经元,散在于Ⅲ~Ⅴ层。2.ChAT免疫反应阳性纤维和终末在灰质各板层内均有分布,其密度依次为前角、中间带、中央灰质和后连合核区、后角。后角Ⅲ层者较密,Ⅰ层次之,而Ⅱ层甚为稀疏。前角诸核团内部及周围有阳性纤维分布,可见大量终扣与大、中型阳性神经元接触,还可见一些轴突末支上生有成串的终扣,围绕着一个细胞的胞体和树突基部,与之形成多个接触点。在中间带各核团间有横行纤维联系。
The morphological characteristies and distribution of the choline acetyltransferase(ChAT)-like immunoreactive neuronal elements in the sacral and coccygeal spinal cord of the cat were studied by avidin-biotin combined immunohistochemical technique.The results were as follows:1.The ChAT-positive neuronsIn the ventral horn,ChAT-positive motoneurons were located in the motor columns.These neurons can be divided into three types:the large multipolar neurons,the medium-sized multipolar neurons and the small motoneurons.The color of anti-ChAT reaction product in the large and medium-sized motoneurons varied from light to moderate brown,but the small motoneurons were heavily stained.In the intermediate zone,three groups of ChAT-positive parasympathetic preganglionic neurons were observed in 3 areas:the intermediolateral nucleus(IL)in the lateral part of laminaⅦ,the intermediomedial nucleus(IM)dorsal-lateral to the central canal,the intercalated nucleus(IC)looated between IL and IM.The somata of these neurons were round,oval,and fusiform in shape.In the laminaⅦ,there were some prominent ChAT medium-sized multipolar neurons named partition cells,scattered among the extended fibers of the parasympathetic preganglionic cells.In the vicinity of the central canal could be observed small CHATpositive cells.In the ventral part of the dorsal commissural nucleus region,a number of ChAT-positive cells were present.They were round,oval and fusiform in shape and small in size(averaging 11.91μm×17.38μm).Occasionally,they were observed in the dorsal part.In addition,some of the ChAT-positive neurons were scattered in laminaeⅢ-Ⅴ.2.The ChAT-positive fibers and terminals.The ChAT-positive punctate structures were observed throughout the sacral and coccygeal spinal cord resembling terminal branch and terminals.The density of the distribution of the punctate structures was arranged,from high to low,in the following order:the ventral horn,intermediate zone,central gray matter and dorsal commissural nucleus region,and the dorsal horn.In the dorsal horn,many of these punctate structures Were concentrated in two laminae,the major one being laminaⅢand a minor one laminaⅠ.But in laminaⅡ,they were rare.In the ventral horn such punctate structures were often closely contacted with large and medium-sized ChAT-positive motoneuronal somata and dendrites.Some axonal terminal branch with varicosities closely surrounded a somata and bases of dendrites,and formed contacts with the same somata and its dendrites.In the intermediate zone,there were many transverse fibers associated among the parasympathetic preganglionic nuclei.
