小白鼠艾氏腹水癌细胞Mg^(++)-ATP酶的细胞化学观察(光学及电子显微镜)

摘要 本实验采用Wachstein-Meisel显示ATP酶的方法,对小白鼠艾氏腹水癌细胞的Mg^(++)-ATP酶进行光学及电子显微镜下的细胞化学观察,见到Mg^(++)-ATP酶集中分布在癌细胞的质膜表面和线粒体内,线粒体内存在的Mg^(++)-ATP酶对戊二醛极为敏感,一经戊二醛固定后,线粒体内存在的Mg^(++)-ATP酶反应消失。但质膜上Mg^(++)-ATP酶的沉淀反应却不受戊二醛的影响,固定后仍可见到强的沉淀反应。L-半胱氨酸可加强质膜上和线粒体内ATP活性酶所产生的阳性反应,而经NaF,PCMB处理后,其阳性的沉淀反应消失。 Activity of Mg^(++)-dependent ATPase in the mitochondria and at the cell surface of Ehrlich ascitic tumor cells of mice was studied by electron-cytochernical and light-histochemical methods during the 5th~8th days after inoculation of Ehrlich ascitic tumor cells.The enzyme,Mg^(++)-dependent ATPase was present in both the Ehrlich ascitic tumor cell surface and mitichondria and the latter was shown to be much sensitive to the glutaldehyde fixatives.So that,the precipitate of lead ion failed to form after the glutaldehyde fixation in the mitochondria,but was still present at the cell surface of the Ehrlich ascitic tumor cells.Moreover,We observed that the activity of the Mg^(++)-ATPase on the cell surface and the mitochondria was increased by the addition of L-Cysteine and inhibited by the addition of NaF,or PCMB to the basal incubating media.
