棉酚对大鼠垂体、肾上腺和下丘脑作用的组织化学观察

摘要 棉酚对男性是具有一定抗生育作用的^([1]),经多方面有关资料的介绍,及根据我们实验材料的观察也得到证实^([2~4])。棉酚的作用环节主要是损伤早期精子细胞和粗线期精母细胞;而对间质细胞可能有轻度影响。睾丸是受垂体促性腺激素的调节,包括促卵泡激素(FSH)和促黄体激素(LH)。这两种激素又受下丘脑分泌的促卵泡素释放激素(FRH)及促黄体素释放激素(LRH)的影响。我们根据棉酚对睾丸作用的情况,认为对垂体和下丘脑有进一步研究和探索的必要。另外,肾上腺是一种高度敏感和应激的器官,对性腺也有密切的关系。因此我们也观察了肾上腺皮质的组织化学变化,作为研究棉酚作用的辅助。 Nineteen male rats(body weight 150~350mg)were divided into five groups,3~4 for each group.Gossypol(30mg/kg/day)was administered in 4 groups for various time du-rations(1,2,3,4 weeks).One group for control and distilled water was given instead.The animals were killed by decapitation.We have observed,the cell bodies of FSH gon-adotropes showed slight enlargment after administration of gossypol for 2-weeks.But LH gonadotropes,TSH cells and acidophils showed no cytological changes.The following changes were observed after administration of gossypol for 3-weeks:The FSH gonadotropes were more hypertrophic,contained a few secretory granules and a large nucleus with a prominent nucleolus.These cells increased in number.The LH gonadotropes showed the same but slighter changes than the FSH gonadotropes.The se-cretory granules of TSH cells decreased in number.No obvious changes were seen in the acidophils.The nerve cells in the paraventricular nucleus were degranulated,and con-tained a large vesicular nucleus.No such changes were observed in neurons of the arcu-ate and supraoptic nuclei.The cells in zone fasciculata and reticularis of adrenal cortex swelled.The lipoid,ascorbic acid and acid phosphatase activity decreased in these cells.Zone glomerulosa and zona intermedia remaided unchanged.
