体外培养肿瘤细胞的核仁内银染颗粒与核仁外移

NUCLEOLAR SILVER-STAINING CRANULES AND NUCLEOLI EXTRUSION IN TUMOUR CELLS IN VITRO

用银染方法显示了体外长期培养食管癌上皮细胞核仁内的银染颗粒,而正常食管上皮细胞在短期培养中用此方法看不到核仁中有明显的颗粒。高温处理后核仁模糊,银染粒不显,经一定时期后又恢复。在此细胞中又观察到核仁外移现象,硫酸锌在一定浓度下可增加核仁外移率,而温度的影响不明显。

Silver stained nucleolar granules were detected in the esophageal cancer cells(ECa 109)in vitro,while they were absent in primary culture of normal esophageal epithelial cells.Immediately after hyperthermic treatment the nucleoli were obscured and the nucleolar granules also disappeared.These granules recovered gradually as the cells were returned to normal culture condition.Analysis of the mitotic stages indicated that the silver-stained structure still persisted at prophase,but hardly seen at metaphase and anaphase,and during telophase a lot of scattered black granules existed over the whole nuclear region.Cases of nucleoli expelled from nuclei to cytoplasm were also seen in these cells.The occurreuce of nucleolar extrusion in cancer cells varied from O to more than(10%).Zinc sulfate increased the rate of nucleolar extrusion,but the effect of hyperthermia was not significant.The relation of nucleolar extrusion and cellular me-tabolism was discussed.
