摘要
目的探讨周期性色氨酸蛋白1(Pwp1)在非小细胞肺癌(NSCLC)的表达与临床病理因素的关系,及对NSCLC恶性表型的影响。方法采用免疫组织化学和Western blot检测NSCLC中Pwp1的表达。采用克隆形成和划痕实验检测Pwp1对NSCLC细胞生物学行为的影响。结果Pwp1在NSCLC阳性表达率为76.5%(62/81),高于癌旁正常组织(21.4%,9/42,P<0.05),与NSCLC低分化(P=0.001)、高TNM分期(P=0.045)和淋巴结转移(P=0.001)相关。Pwp1在NSCLC细胞系高表达,干扰Pwp1明显抑制NSCLC细胞系的增殖和迁移。结论Pwp1是NSCLC治疗的潜在靶点,干扰Pwp1表达抑制NSCLC细胞的恶性表型。
Objective To explore the relationship between the expression of periodic tryptophan protein(Pwp1)and clinicopathological factors in non-small cell lung cancer(NSCLC),and the effect on the biological function of NSCLC cells.Methods Immunohistochemistry and Western blot were performed to detect the expression of Pwp1 in NSCLC.Colony formation and wound healing were performed to test the proliferation and migration of NSCLC.Results The positive rate of Pwp1 was significantly higher in NSCLC cells(76.5%,62/81)than in normal lung tissues(21.4%,9/42,P<0.05),positively related to low differentiation(P=0.001),high TNM stage(P=0.045)and lymph node metastasis(P=0.001),and also Pwp1 was highly expressed in NSCLC cell lines.Transfection of Pwp1-siRNA downregulated the proliferation and migration of NSCLC cell lines significantly.Conclusion Pwp1 is a potential target for the treatment of NSCLC,interferon of Pwp1 inhibited the malignant phenotype of NSCLC cells.
