褐藻素对肝癌HepG2细胞增殖、凋亡与侵袭的影响及可能机制

Effects of fucoxanthin on proliferation,apoptosis and invasion of HepG2 cells and its possible mechanism

目的探讨褐藻素对肝癌HepG2细胞增殖、凋亡和侵袭的影响。方法体外培养人肝癌细胞HepG2,将其分为对照组(DMSO)、15、30、45μmol/L褐藻素组。各组细胞以不同浓度药物处理48h后,MTT实验与平板克隆实验检测HepG2细胞增殖能力变化;流式细胞术检测HepG2细胞的细胞周期和凋亡情况;Transwell实验检测HepG2细胞侵袭能力变化;Western blot方法检测HepG2细胞p-Akt、Cyclin D1、基质金属蛋白酶2/9(MMP2/9)的表达。结果与对照组比较,不同浓度褐藻素均能抑制HepG2细胞增殖、阻滞细胞周期、诱导凋亡、降低侵袭能力(P<0.05)。Western blot结果显示,褐藻素能够降低HepG2细胞中p-Akt、Cyclin D1、MMP2/9的表达(P<0.05)。结论褐藻素能够抑制HepG2细胞增殖与侵袭能力,阻滞细胞周期并诱导凋亡。

Objective To investigate the effects of fucoxanthin on proliferation,apoptosis and invasion of Hepatoma HepG2 cells.Methods HepG2 cells were cultured in vitro and divided into control group(DMSO),15,30 and 45μmol/L fucoxanthin groups.HepG2 cells were treated with different concentrations of drugs for 48h.Proliferation of HepG2 cells was detected by MTT assay and plate cloning assay.The cell cycle and apoptosis of HepG2 cells were detected by flow cytometry.The invasion ability of HepG2 cells was detected by Transwell assay.The expressions of p-Akt,Cyclin D1 and matrix metalloproteinases 2/9(MMP2/9)in HepG2 cells were detected by Western blot.Results Compared with the control group,different concentrations of fucoxanthin could inhibit the proliferation,block the cell cycle,induce apoptosis and decrease invasion ability of HepG2 cells(P<0.05).The results of Western blot showed that fucoxanthin could inhibit the expressions of p-Akt,Cyclin D1 and MMP2/9 in HepG2 cells(P<0.05).Conclusion Fucoxanthin can inhibit the proliferation and invasion of HepG2 cells,block cell cycle and induce apoptosis.
