MicroRNA-193b通过靶向调控SOX3基因对宫颈癌Hela细胞增殖和凋亡的影响

MicroRNA-193b regulates the proliferation and apoptosis of cervical cancer Hela cell by targeting the SOX3 gene

摘要 目的探讨miRNA-193b对宫颈癌细胞Hela增殖、凋亡的作用机制。方法将对数生长期的宫颈癌Hela细胞随机分为miRNA-193b模拟物(miRNA-193b-mimics)组、miRNA-193b模拟物阴性对照(miRNA-193b-mimics-NC)组及对照(Control)组。Real-time PCR检测三组细胞miRNA-193b表达水平;CCK-8法检测Hela细胞的细胞活力;EDU染色和细胞克隆实验检测Hela细胞的增殖能力;流式细胞仪检测细胞凋亡情况;Real-time PCR、Western blot检测SOX3mRNA和蛋白的相对表达水平;双荧光素酶活性实验验证miRNA-193b对SOX3的靶向作用。结果与Control组和miRNA-193b-mimics-NC相比,miRNA-193b-mimics组细胞miRNA-193b表达水平明显增加(P<0.05);细胞活力与增殖能力明显下降,细胞凋亡比例明显升高(P<0.05);SOX3 mRNA和蛋白表达水平明显降低(P<0.05);双荧光素酶报告基因结果显示SOX3为miRNA-193b的靶基因。结论MicroRNA-193b调控宫颈癌Hela细胞增殖和凋亡的作用机制可能与靶向调控SOX3表达有关。 Objective To investigate the mechanism of miRNA-193b on the proliferation and apoptosis of cervical cancer Hela cells.Methods Hela cells in logarithmic growth phase were divided into miRNA-193b-mimics,miRNA-193b-mimics-NC and control group.The level of miRNA-193b was detected by Real-time PCR.The cell viability of Hela cells was detected by CCK-8 method.The proliferation ability of Hela cells was detected by EDU staining and cell cloning assay.Apoptosis of cells was detected by flow cytometry.The relative expression of SOX3 mRNA and protein was detected by Realtime PCR and Western blot.Double luciferase activity assay verified the targeting effect of miRNA-193b on SOX3.Results The expression of miRNA-193b of cells in the miRNA-193b-mimics group was significantly increased(P<0.05).Cell viability and proliferative ability of cells in the miRNA-193b-mimics group were significantly decreased,while apoptosis ratio was significantly increased(P<0.05).The levels of mRNA and protein of SOX3 were significantly decreased(P<0.05).Dual luciferase reporter gene results showed that SOX3 was the target gene of miRNA-193b.Conclusion The mechanism of microRNA-193b regulating the proliferation and apoptosis of cervical cancer Hela cells may be related to the targeted regulation of SOX3 expression.
