PDGFRβ对人脐静脉血管内皮细胞增殖、凋亡和成管的影响

The effect of PDGFRβon proliferation,apoptosis and tube formation ability of human umbilical vein endothelial cells

摘要 目的探索PDGFRβ在高糖培养的血管内皮细胞中的表达情况及其对血管内皮细胞增殖、凋亡及成管的影响和机制。方法在体外培养人脐静脉血管内皮细胞,构建高糖模型,转染si-RNA敲减PDGFRβ,将细胞分为Control组、35mM组、si-PDGFRβ+35mM组。qRT-PCR方法检测各组细胞PDGFRβ的mRNA表达情况;CCK8方法检测各组细胞增殖能力的变化;流式细胞术检测细胞凋亡情况;血管形成实验检测成管能力的变化。结果与Control组相比,35mM组中PDGFRβ的mRNA表达升高(P<0.05)。在35mM组的细胞中敲低PDGFRβ后,细胞的增殖能力有所恢复,凋亡情况有所减少,成管能力降低(P<0.05)。结论PDGFRβ在高糖培养的人脐静脉内皮细胞中高表达,敲减PDGFRβ可以恢复其增殖能力、减轻凋亡情况、降低成管能力。 Objective To explore the expression of PDGFRβin vascular endothelial cells cultured with high glucose and its effect on proliferation,apoptosis and tubular formation of vascular endothelial cells.Methods Human umbilical vein endothelial cells were cultured in vitro to construct high glucose model and transfected with si-RNA knockdown PDGFRβ.The cells were divided into Control group,35mM group and si-PDGFRβ+35mM group.The expression of PDGFRβmRNA was detected by qRT-PCR.The proliferation ability of cells was detected by CCK8 method.Apoptosis of cells was detected by flow cytometry.Ability of tubefor ming was detected by angiogenesis.Results Compared with the control group,the expression of PDGFRβmRNA in the 35mM group was increased(P<0.05).After knocking down the PDGFRβin the 35mM group,the proliferation ability of cells was restored,the apoptosis of cells was reduced,and the ability of tube formation was reduced(P<0.05).Conclusion PDGFRβis highly expressed in human umbilical vein endothelial cells cultured with high glucose.Knocking down PDGFRβcan restore its proliferative ability,alleviate apoptosis and reduce its tubefor ming ability.
