独活寄生汤对肩周炎模型兔的治疗作用及可能机制 被引量:2

Therapeutic effect and possible mechanism of Duhuo Jisheng Decoction on the rabbit model of periarthritis of shoulder

摘要 目的探究独活寄生汤对肩周炎模型兔的治疗作用及作用机制。方法40只健康雄性新西兰兔随机挑选10只作为空白组(Control组),其余30只通过持续机械劳损加冰敷的方式制造肩周炎家兔模型,将造模成功的30只家兔再随机分为模型组(Model组)、独活寄生汤低剂量组(DHJSD-L组)、独活寄生汤高剂量组(DHJSD-H组),每组10只。ELISA检测家兔血清炎症因子IL-1β、IL-18、TNF-α及疼痛有关因子5-HT、PGE_(2)的表达情况;免疫荧光法检测NF-κB蛋白表达;Western blot检测家兔肩关节滑膜组织NF-κB、GSDMD-C、GSDMD-N、NLRP3、ASC、pro-Caspase-1及pro-IL-1β蛋白表达水平;qRT-PCR检测家兔肩关节滑膜组织NLRP3、ASC、Caspase-1与IL-1βmRNA表达情况。结果与模型组相比,独活寄生汤可明显抑制肩周炎模型兔IL-1β、IL-18、TNF-α、5-HT及PGE_(2)的表达;显著减弱NF-κB荧光强度;下调肩周炎模型兔肩关节滑膜组织中NF-κB、GSDMD-C、GSDMD-N、NLRP3、ASC、pro-Caspase-1及pro-IL-1β蛋白的表达,下调肩关节滑膜组织NLRP3、ASC、Caspase-1与IL-1β的mRNA表达水平。结论独活寄生汤可改善肩周炎的症状并缓解疼痛,其治疗作用可能是通过调控NF-κB/NLRP3/Caspase-1通路来实现的。 Objective To explore the therapeutic effect and possible mechanism of Duhuo Jisheng Decoction on the rabbit model of periarthritis of shoulder.Methods Forty healthy male New Zealand rabbits were randomly selected as control group(10 rabbits),and the other 30 rabbits were treated with continued strain plus ice compress to create a rabbit model of scapulohumeral periarthritis.Thirty rabbits with successful modeling were randomly divided into model group,low-dose Duhuo Jisheng Decoction group(DHJSD-L group)and high-dose Duhuo Jisheng Decoction group(DHJSD-H group),with 10 rabbits in each group.The levels of IL-1β,IL-18,TNF-α,5-HT and PGE_(2)in serum of rabbits were detected by ELISA.The expression of NF-κB was detected by immunofluorescence staining.The expressions of NF-κB,GSDMD-C,GSDMD-N,NLRP3,ASC,pro-Caspase-1 and pro-IL-1βin rabbit shoulder synovium were detected by Western blot.The mRNA levels of NLRP3,ASC,Caspase-1 and IL-1βin rabbit shoulder synovium were detected by qRT-PCR.Results Compared with the model group,Duhuo Jisheng Decoction can significantly inhibit the expressions of IL-1β,IL-18,TNF-α,5-HT and PGE_(2)in rabbits with shoulder periarthritis model.The fluorescence intensity of NF-κB was decreased significantly.The expressions of NF-κB,GSDMD-C,GSDMD-N,NLRP3,ASC,pro-Caspase-1 and pro-IL-1βwere down-regulated,and the mRNA levels of NLRP3,ASC,Caspase-1 and IL-1βwere also significantly down-regulated in periarthritis of scapulohumeral periarthritis rabbit models.Conclusion Duhuo Jisheng Decoction can improve the symptoms and relieve pain of scapulohumeral periarthritis,and its therapeutic effect may be realized by regulating NF-κB/NLRP3/Caspase-1 pathway.
