摘要
目的探索长链非编码RNA PVT1在卵巢癌细胞中的表达及其对卵巢癌细胞增殖和侵袭的影响。方法在人卵巢癌细胞系和人卵巢上皮细胞系中,用qRT-PCR检测PVT1和miR-145的表达。在人卵巢癌SKOV3细胞中转染PVT1干扰序列,下调PVT1表达,应用qRT-PCR检测干扰PVT1表达对miR-145表达的影响;MTT法检测干扰PVT1表达对SKOV3细胞增殖的影响;Transwell实验检测干扰PVT1表达对SKOV3细胞侵袭能力的影响。生物学网站分析及双荧光素酶报告基因实验验证PVT1和miR-145的靶向关系。结果与人卵巢上皮细胞系HOSE相比,PVT1在卵巢癌细胞SKOV3中表达升高,miR-145在卵巢癌细胞SKOV3中表达下降。转染si-PVT1可以增加卵巢癌细胞中miR-145的表达。与对照组相比,si-PVT1组卵巢癌细胞的增殖、侵袭能力下降(P<0.05)。生物信息学和荧光素酶分析表明PVT1可以海绵吸附miR-145。结论lncRNA PVT1在卵巢癌细胞系中高表达,抑制lncRNA PVT1可以抑制卵巢癌细胞SKOV3的增殖及侵袭,机制可能与影响miR-145表达有关。
Objective To explore the expression of long non-coding RNA PVT1 in ovarian cancer cells and its effect on the proliferation and invasion of ovarian cancer cells.Methods The expressions of PVT1 and miR-145 in human ovarian cancer cell line and normal human ovarian epithelial cell line were detected by qRT-PCR.SKOV3 cells were transfected with PVT1 interference sequence and down-regulated PVT1 expression.qRT-PCR was used to detect the expression of miR-145.MTT assay was used to detect the proliferation of SKOV3 cells.Transwell assay was used to detect the invasion ability of SKOV3 cells.The targeting relationship between PVT1 and miR-145 was verified by biological website analysis and double luciferase reporter assay.Results Compared with the human ovarian epithelial cell line HOSE,the expression of PVT1 was increased in SKOV3 cells,and the expression of miR-145 was decreased in SKOV3 cells.Transfection of si-PVT1 can increase the expression of miR-145 in ovarian cancer cells.Compared with the control group,the proliferation and invasion ability of ovarian cancer cells in si-PVT1 group were decreased(P<0.05).Bioinformatics and luciferase analysis showed that PVT1 could sponge adsorb miR-145.Conclusion lncRNA PVT1 is highly expressed in ovarian cancer cell lines.Inhibition of lncRNA PVT1 can inhibit the proliferation and invasion of SKOV3 cells,and the mechanism may be related to affecting the expression of miR-145.
