摘要
目的研究尪痹胶囊抗碘乙酸钠(MIA)所致大鼠膝关节炎软骨降解的作用。方法48只SD大鼠,随机分为空白组、模型组、氨基葡萄糖阳性药组、尪痹胶囊低剂量组、尪痹胶囊中剂量组、尪痹胶囊高剂量组,使用向大鼠左侧膝关节腔缓慢注射MIA的方法来建立膝关节炎软骨降解模型。ELISA法检测各组大鼠血清中TNF-α、IL-1β表达水平;取大鼠右侧膝关节进行大鼠膝关节直径测定和关节评分并且进行Micro-CT扫描,获得骨结构参数结果;取右侧膝关节软骨组织,进行苏木素-伊红(HE)染色与番红-O-固绿染色。结果与模型组相比,经低、中、高剂量尪痹胶囊治疗后,关节肿胀得到消退,软骨表面光泽度、缺损情况得到改善,骨体积分数的上升与骨小梁数量的下降得到抑制,IL-1β和TNF-α的浓度出现下降(P<0.05)。结论尪痹胶囊能有效改善MIA所致大鼠膝关节炎软骨降解。
Objective To study the effect of Wangbi Capsule on sodium iodoacetate(MIA)-induced cartilage degradation of knee arthritis.Methods Forty-eight Sprague-Dawley rats were randomly divided into blank group model group,glucosamine positive drug group,Wangbi Capsule low-dose group,Wangbi Capsule medium-dose group and Wangbi Capsule high-dose group.The cartilage degradation model was established by slowly injecting MIA into the left knee joint cavity of the rat.The levels of TNF-αand IL-1βin serum were detected by ELISA.The diameter and joint score of the right knee joint of the rat were measured and Micro-CT scan was performed to obtain the results of bone structure parameters.The cartilage tissue of the right knee joint was taken and stained with hematoxylin-eosin(HE)and saffranineO-solid green.Results Compared with the model group,the joint swelling and cartilage surface glossiness and defects were improved after receiving low,medium and high doses of WP treatment,the increase of bone volume fraction and decrease of bone trabecular number were inhibited,and the concentrations of IL-1βand TNF-αwere decreased(P<0.05).Conclusion Wangbi Capsule can effectively improve the cartilage degradation of MIA-induced osteoarthritis in rats.
