干扰ERCC6L增强非小细胞肺癌细胞放疗敏感性

Interference with ERCC6L enhances radiosensitivity of non-small cell lung cancer cells

摘要 目的探讨切除修复交叉互补酶6L(ERCC6L)在非小细胞肺癌中的表达及其与放射敏感性的关系。方法Western blot和免疫荧光验证ERCC6L在肺癌细胞系中的表达水平及亚细胞定位;Western blot检测16对癌组织及癌旁组织中ERCC6L的表达;免疫组化的方法检测肺癌和X射线照射后肺癌组织中ERCC6L的表达;用ERCC6L特异性siRNA干扰其在A549细胞中的表达,用磷酸化-γH2AX荧光焦点检测DNA双链断裂损伤的修复情况。结果ERCC6L在肺癌组织的中表达水平高于癌旁组织,在A549细胞中干扰ERCC6L并经过X射线照射后,抑制DNA双链断裂损伤的修复能力。结论干扰ERCC6L可以增强非小细胞肺癌的放射敏感性。 Objective To investigate the expression of excision repair cross complementing enzyme 6L(ERCC6L)in non-small cell lung cancer(NSCLC)and its influence on radiosensitivity of NSCLC.Methods Western blot and immunofluorescence were performed to verify the expression level and subcellular localization of ERCC6L in lung cancer cell lines.The expression of ERCC6L in 16 pairs of lung cancer tissues and adjacent tissues was examined by Western blot.Immunohistochemistry was used to detect the expression of ERCC6L in lung cancer and lung cancer tissues which irradiated by X-ray.ERCC6L-specific siRNA was used to interfere with its expression in A549 cells.Phosphorylation-γH2AX foci numbers were used to detect the repair of DNA double-strand breakage.Results The expression of ERCC6L was higher in lung cancer tissues than in adjacent tissues(P<0.05).After interfering with ERCC6L by siRNA in A549 cells and irradiated by X-ray,DNA double strand breakage repair ability was inhibited.Conclusion Inhibiting ERCC6L enhances the radiosensitivity of NSCLC.
