海兰白蛋鸡法氏囊中T淋巴细胞的发育规律

摘要 为研究不同日龄海兰白蛋鸡法氏囊中T淋巴细胞发育规律,试验分别选择15,18,20胚龄和出生后1,4,7,10,14,21,35,56日龄海兰白鸡胚和雏鸡进行采样,将所采集到的法氏囊样本制备超薄冰冻切片,通过免疫组织化学方法结合生物显微图像分析技术,研究不同发育阶段鸡法氏囊中的CD3+、CD4+和CD8+T淋巴细胞的出现、迁移、组织定位和分布。结果表明:15胚龄鸡法氏囊首次检测到CD3+和CD4+T淋巴细胞,其散在的分布于法氏囊淋巴滤泡中,在4日龄以后同时分布于淋巴滤泡皮质和髓质交界的结缔组织和小梁中。18胚龄鸡法氏囊中首次检测到CD8+T淋巴细胞,1日龄后CD8+T淋巴细胞分布规律与CD3+和CD4+T淋巴细胞相似,开始为散在地分布于淋巴滤泡中,之后逐渐向皮质边缘和皮质间小梁处靠近,到56日龄时,CD8+T淋巴细胞主要分布于皮质间小梁处。这说明,随着日龄增长,鸡法氏囊中T淋巴细胞及其亚群的分布和数量呈现日龄相关性变化,这对探讨蛋鸡在胚胎时期以及出壳初期的法氏囊的细胞免疫状态具有重要意义,同时也为鸡群适时免疫和疾病防治等相关研究提供一定参考依据。 The purpose of this experiment is to study the development of T lymphocytes in the bursa of Fabricius in Hailan white layers of different ages.The experiment selected 15,18 and 20 embryonic ages and 1,4,7,10,14,21,35 and 56-day-old Hylan white chicken embryos and chicks for sampling.The collected samples of the bursa of Fabricius were prepared into ultra-thin frozen sections.The appearance,migration,tissue localization and distribution of CD3~+,CD4~+and CD8~+T lymphocytes in the chicken bursa of Fabricius at different developmental stages were studied by immunohistochemistry combined with biological microscopic image analysis technology.The results showed that CD3~+and CD4~+T lymphocytes were detected for the first time in the bursa of fabric at 15 embryonic age chickens.They were scattered in the lymphoid follicles of the bursa of fabric,and distributed in the connective tissue and trabeculaeat of lymphoid follicular cortex and medulla simultaneously after 4 days of age.CD8~+T lymphocytes were detected for the first time in the bursa of 18 embryonic chickens.After 1 day of age,the distribution of CD8~+T lymphocytes was similar to CD3~+and CD4~+T lymphocytes.It began to be scattered in lymph follicles,and then gradually approached the cortex and the intercortical trabecular.By 56 days of age,CD8~+T lymphocytes were mainly distributed in the intercortical trabecular.The results showed that with the increase of age,the distribution and number of T lymphocytes and their subgroups in the bursa of chickens showed age-related changes,which is of great significance to study the cellular immune status of the bursa during the embryonic period and the early stage of hatching.At the same time,it provides a certain reference for timely immunization and disease prevention and control of chickens.
