聚乙二醇胁迫下黄腐酸对玉米基因组DNA胞嘧啶甲基化的影响 被引量:1

Effects of Fulvic Acid on Cytosine Methylation of Maize Genomic DNA under PEG Stress

摘要 用不同浓度的黄腐酸浸种处理玉米自交系B73种子,2叶1心时用10%PEG-6000模拟干旱胁迫,3叶期测定干旱相关生理指标,采用甲基化敏感扩增多态性(MSAP)技术分析DNA甲基化模式及水平。结果表明:各处理组的甲基化敏感扩增多态性分别为74.83%、73.05%、76.50%;各处理组的甲基化敏感扩增单态性分别为25.17%、26.95%、23.50%。各试验组的总甲基化率分别为75.14%、77.02%、74.57%;半甲基化率分别为18.01%、23.39%、18.66%;全甲基化率分别为57.95%、57.13%、53.63%、55.91%。 The aim of this study is to reveal the effects of fulvic acid treatment on cytosine methylation of maize genomic DNA under polyethylene glycol stress.The seeds of maize inbred line B73 were soaked with fulvic acid at different concentrations.During the two-leaf and one-core stage,10%PEG-6000 was used to simulate drought stress,and drought related physiological indexes were measured in the three-leaf stage.The DNA methylation patterns and levels were analyzed by methylation sensitive amplified polymorphism(MSAP).The results show that:the polymorphisms of methylation-sensitive amplification in each treatment group are 74.83%,73.05%and 76.50%,respectively,and the monomorphisms of methylation-sensitive amplification in each treatment group are25.17%,26.95%and 23.50%,respectively;the total methylation rates in each experimental group are 75.14%,77.02%and 74.57%,respectively,the hemimethylation rates are 18.01%,23.39%and 18.66%,respectively,and the permethylation rates are 57.95%,57.13%,53.63%and 55.91%,respectively.
