葡萄糖促进猪瘟病毒增殖的研究

摘要 病毒依赖宿主细胞代谢以获取复制所需的能量和生物合成分子。为探索葡萄糖和谷氨酰胺在猪瘟病毒增殖中的作用,将剥夺了葡萄糖和谷氨酰胺或加有不同浓度葡萄糖和葡萄糖类似物2-脱氧葡萄糖(2-Deoxy-D-glucose,2-DG)的培养基用于猪瘟病毒感染细胞的培养,感染后24 h收集样品,进行病毒滴度和基因组拷贝数测定,以及病毒蛋白表达的鉴定。结果表明:与正常对照相比,谷氨酰胺剥夺对病毒滴度没有显著影响,而葡萄糖剥夺时病毒滴度和基因组拷贝数分别降低30倍和20倍,同时病毒滴度与葡萄糖的浓度呈正相关。表明葡萄糖是猪瘟病毒增殖的必需营养物质。2-DG处理也能显著抑制猪瘟病毒的增殖。综上,葡萄糖对猪瘟病毒增殖至关重要,缺少葡萄糖则猪瘟病毒无法正常复制,而谷氨酰胺对猪瘟病毒增殖并无影响。 Viruses rely on host cellular metabolism to provide energy and biosynthetic building blocks required for its replication. To explore the role of glucose and glutamine in the replication of classical swine fever virus(CSFV), cell culture medium, which is deprived of glucose and glutamine or contains different concentrations of glucose or 2-Deoxy-D-glucose(2-DG), was used for culturing CSFV-infected cells, infected cells were harvested at 24 h postinfection and then subjected to detect viral titer, genomic copies and expression levels of viral protein. The results showed that deprivation of glucose caused 30-fold and 20-fold reduction of infectious virus titer and genomic copies, respectively, viral replication levels were positively related to the concentrations of glucose, but deprivation of glutamine had no significant impact on CSFV replication. Furthermore, 2-DG could also significantly inhibit the replication of CSFV. To conclude, glucose is critical for CSFV replication rather than glutamine, which will be useful for elucidating the relation between CSFV replication and cellular metabolism in future.
