酸枣仁汤对慢性睡眠剥夺大鼠的药效学研究 被引量:3

Pharmacodynamic Study of Suanzaoren Decoction on Rats with Chronic Sleep Deprivation

摘要 目的建立睡眠剥夺大鼠模型,探讨酸枣仁汤对睡眠剥夺大鼠的药效。方法选用健康成年雄性SD大鼠随机分为正常组、模型组、褪黑素组、酸枣汤组。参照文献建立慢性睡眠剥夺大鼠模型。各组给予相应处理后,采用旷场试验比较各组总活动距离、总穿格数、直立次数和直立时间;观察并记录各组睡眠潜伏期和睡眠时间;采用酶联免疫吸附法测定大鼠下丘脑组织中谷氨酸(Glu)和γ氨基丁酸(GABA)的含量。结果与正常组相比,模型组大鼠总活动距离、总穿格数、站立次数、站立时间均有显著下降(P<0.01);与模型组相比,褪黑素组、酸枣仁汤组大鼠总活动距离、总穿格数、站立次数、站立时间均有显著增加(P<0.01)。与正常组相比模型组睡眠潜伏期增加,睡眠时间减少(P<0.01);与模型组相比,褪黑素组、酸枣仁汤组大鼠睡眠潜伏期减少,睡眠时间增加(P<0.01);褪黑素组与酸枣仁汤组大鼠睡眠潜伏期和睡眠时间差异不显著(P>0.05)。与正常组相比,模型组Glu增高,GABA下降,Glu/GABA升高(P<0.01)与模型组相比,褪黑素组、酸枣仁组大鼠Glu下降,GABA增高,Glu/GABA下降(P<0.01);褪黑素组与酸枣仁汤组大鼠Glu、GABA含量和Glu/GABA差异不显著(P>0.05)。结论酸枣仁汤对睡眠剥夺大鼠的焦虑和睡眠有明显的保护作用,可能通过降低下丘脑组织中Glu的含量、增加GABA的含量恢复脑内神经元兴奋和抑制的平衡状态发挥作用。 Objective To establish a rat model of sleep deprivation and explore the efficacy of Suanzaoren Decoction on sleep deprived rats.Methods Healthy adult male SD rats were randomly divided into normal group,model group,melatonin group and Suanzao Decoction group.A rat model of chronic sleep deprivation was established according to literature.After each group was given corresponding treatment,the open field test was used to compare the total movement distance,total number of grids,upright times and upright time of each group.The sleep latency and sleep time of each group was observed and recorded.Enzyme-linked immunosorbent assay(ELISA)was used to determine the content of glutamic acid(Glu)and y-aminobutyric acid(GABA)in rat hypothalamus.Results Compared with normal group,the total movement distance,total number of grids,standing times,and standing time of model group were significantly decreased(P<0.01).Compared with model group,the total movement distance,total number of grids,standing times,and standing time of rats increased significantly in melatonin group and Suanzaoren decoction group(P<0.01);compared with normal group,the sleep latency of model group increased and the sleep time decreased(P<0.01);compared with model group,the sleep latency of melatonin group and Suanzaoren Decoction group decreased,and the sleep time increased(P<0.011),there was no significant difference in sleep latency and sleep time between melatonin group and Suanzaoren Decoction group(P>0.05);compared with normal group,Glu,GABA and Glu/GABA increased in model group(P<0.01);compared with model group,Glu decreased,GABA and Glu/GABA increased in melatonin group and Suanzaoren Decoction group decreased(P<0.01),there were no significant differences in Glu,GABA content and Glu/GABA between melatonin group and Suanzaoren Decoction group(P>0.05).Conclusion Suanzaoren Decoction has obvious protective effect on anxiety and sleep of sleep deprivation rats,which may play a role by reducing the content of Glu in hypothalamus and increasing the content of GABA to restore the balance state of excitation and inhibition of neurons in the brain.
