川楝树提取物对人乳腺癌细胞增殖、凋亡、迁移及细胞周期分布的影响 被引量:1

Effects of Toosendani Extract on Proliferation,Apoptosis,Migration and Cell Cycle Distribution of Human Breast Cancer Cells

摘要 目的研究川楝树提取物对人乳腺癌细胞增殖、凋亡、迁移以及细胞周期分布的影响。方法将人乳腺癌MCF-7细胞分为空白组和川楝低、中、高浓度组,采用MTT法和流式细胞术检测细胞不同时间段乳腺癌细胞的增殖和凋亡情况,Transwell实验检测细胞迁移情况,流式细胞仪检测细胞周期分布,Western blot检测ICAM-1、MMP-2、MMP-9的表达。结果川楝低、中、高浓度组乳腺癌细胞在不同时间段的增殖率、凋亡率均显著低于和高于空白组,且随着浓度的升高作用效果更加明显(P<0.05,P<0.01);川楝低、中、高浓度组乳腺癌细胞的细胞迁移数量相比空白组均显著减少,且细胞迁移个数随着浓度的升高而明显减少;川楝树提取物浓度越高,乳腺癌细胞处于G0/G1和S周期的细胞比例显著降低,处于G2/M周期的细胞比例明显上升(P<0.01);川楝低、中、高浓度组ICAM-1、MMP-2、MMP-9的表达显著低于空白组,高、中浓度组ICAM-1、MMP-2、MMP-9的表达明显低于低浓度组(P<0.01)。结论川楝树提取物能够有效抑制乳腺癌细胞的增殖和迁移,促进癌细胞的凋亡,通过改变癌细胞的周期分布降低癌细胞的增长,能力呈浓度依赖性。 Objective To study the effects of toosendani extract on proliferation,apoptosis,migration and cell cycle distribution of human breast cancer cells. Methods Human breast cancer MCF-7 cells were divided into blank group,toosendani extract low,medium and high concentration groups. PBS solution and sterile physiological saline were added into the blank group for cell culture. MTT and flow cytometry were used to detect cell proliferation and apoptosis at different time periods,Transwell assay was used to detect cell migration,flow cytometry was used to detect cell cycle distribution,and Intercellular cell adhesion molecule-1(ICAM-1),Matrix metalloproteinase-2(MMP-2),Matrix metalloproteinase-9(MMP-9)were detected by Western blot. Results The proliferation rate of breast cancer cells in toosendani extract low,medium and high concentration groups was significantly lower than that in blank group at different time periods,and the apoptosis rate was significantly higher than that in blank group,and the effect was more obvious with the increase of concentration( P<0.05, P<0.01). The number of cell migration of breast cancer cells in low,medium and high concentration groups was significantly reduced compared with blank group,and the number of cell migration was significantly reduced with the increase of the toosendani extract concentration( P<0.05). The higher the toosendani extract,the lower the proportion of breast cancer cells in G0/G1 and S cycle and the higher the proportion of breast cancer cells in G2/M cycle( P<0.01). The expressions of ICAM-1,MMP-2 and MMP-9 in toosendani extract low,medium and high concentration groups were significantly lower than those in blank group,and the expressions of ICAM-1,MMP-2 and MMP-9 in high and medium concentration groups were significantly lower than those in low concentration group( P<0.01). Conclusion The extract can effectively inhibit the proliferation and migration of breast cancer cells,promote the apoptosis of cancer cells,and reduce the growth of cancer cells by changing the periodic distribution of cancer cells,and the ability is concentration dependent.
