新型微创小鼠主动脉弓缩窄模型的建立

Establishment of a novel minimally invasive transverse aortic constriction model in mice

摘要 目的建立一种新型微创小鼠主动脉弓缩窄(transverse aortic constriction,TAC)模型。方法将18只C57BL/6雄性小鼠随机分为对照组(n=9)和新型微创主动脉弓缩窄术(novel minimally invasive transverse aortic constriction,NMI-TAC)组(n=9)。对照组不做处理,NMI-TAC组进行新型微创TAC术。记录平均手术时间、术中及术后4周生存率。术后第4周采用超声检测心功能,HE染色观察心肌肥厚程度,Masson染色观察心肌纤维化程度。结果NMI-TAC组手术时间约为(13.28±2.12)min,术中无小鼠死亡,术后1周死亡1只,生存率为88.89%。与对照组比较,NMI-TAC组心脏超声显示心室壁增厚,左室射血分数(left ventricular ejection fractions,LVEF)、左室缩短分数(left ventricular fractional shortening,LVFS)增加(P<0.05)。HE染色显示NMI-TAC组心内膜下心肌细胞肥大。Masson染色显示NMI-TAC组心肌间质和血管周围纤维化程度明显增加。结论新型微创TAC术能够成功构建小鼠主动脉弓缩窄模型,并诱导心肌肥厚。 Objective To establish a novel minimally invasive transverse aortic constriction(TAC)model in mice.Methods Eighteen C57 BL/6 male mice were randomly divided into control group(n=9)and novel minimally invasive transverse aortic constriction(NMI-TAC)group(n=9).No treatment was given to the control group,and the NMI-TAC group was operated with NMI-TAC surgery.Mean operation time and overall survival rate during operation and 4 weeks after operation were recorded.Echocardiography was used to detect cardiac function in the 4 th week after TAC surgery,HE staining was used to observe cardiac hypertrophy degree,Masson staining was used to observe cardiac fibrosis degree.Results The average operation time using NMI-TAC surgery was(13.28±2.12)min.There was no intraoperative death and 1 postoperative death.The survival rate was 88.89%.Compared with the control group,echocardiography showed ventricular wall thickening,and the left ventricular ejection fractions(LVEF)and the left ventricular fractional shortening(LVFS)were both increased in NMI-TAC group(P<0.05).HE staining showed subendocardial cardiomyocyte hypertrophy.Masson staining showed that the degree of myocardial interstitial and perivascular fibrosis increased significantly.Conclusion The novel minimally invasive TAC surgery could successfully construct the TAC model and induce cardiac hypertrophy in mice.
