肌源干细胞移植修复关节软骨损伤的实验研究

Experimental study on repair of cartilage tissue damage by muscle derived stem cell transplantation

摘要 目的利用小鼠关节软骨损伤模型,确定肌源干细胞具有一定的关节软骨损伤修复作用。方法选取72只6~8周龄的雄性BALB/C裸鼠,随机分为2组,采用平头钻头钻孔,造成直径1 mm,深度0.5 mm的关节软骨缺损模型,A组将120μL的MDSCs细胞悬液注射至关节软骨组织损伤处后缝合好关节囊,闭合创口,B组注入DMEM/F12,然后分别于术后4、8、12周后取各组12只裸鼠处死,取双侧膝关节观察关节囊是否渗出、滑膜增生,修复组织的高低、平整程度、连续性、光泽度及是否有裂隙。对各组关节骨外科专家采用盲法按照国际软骨修复学会ICRS评分标准评分后进行统计学分析。结果A组修复组织与周围软骨高度、色泽一致,已难以区分边界,B组修复不平整,中央凹陷,修复组织自身出现龟裂,与周围软骨出现裂隙。结论将肌源干细胞植入小鼠损伤模型中可以更加良好快速地帮助小鼠关节软骨损伤模型的修复。 Objective To determine if the muscle derived stem cells have a certain repairing effect on arthrosis cartilage damage in the mouse arthrosis cartilage injury model.Methods Seventy-two male BALB/C nude mice aged 6~8 weeks were randomly divided into two groups and drilled with a flat drill to create a arthrosis cartilage defect model with a diameter of 1 mm and a depth of 0.5 mm.Group A was injected with 120μL of MDSCs cell suspension into the arthrosis cartilage tissue injury site,the joint capsule was sutured and the wound was closed,and the B group was injected with DMEM/F12.Then,4,8 and 12 weeks after operation,12 nude mice in each group were sacrificed,and the joint capsule exudes,synovial hyperplasia,the repair height,flatness,continuity,gloss and crack of the tissue of bilateral knee joints were observed.Statistical analysis was performed on each group by articular orthopedic surgeons using the blind method according to the International Cartilage Restoration Society ICRS score.Results The repaired tissue of group A was consistent with the height and color of the surrounding cartilage.It was difficult to distinguish the boundary.But that of group B was uneven,the central depression,the repaired tissue cracked itself,and the surrounding cartilage cracked.Conclusion The muscle derived stem cells implanted into mice can help the repair of the arthrosis cartilage injury model much more quickly.
