蛇床子素对微重力下Th1/Th2细胞亚群平衡的免疫调节作用 被引量:3

Immunomodulatory effect of osthole on the balance of Th1 and Th2 cell under microgravity

摘要 目的研究蛇床子素在微重力条件下对Th1/Th2细胞亚群平衡的调节作用。方法应用后肢悬吊法建立SD大鼠微重力模型。造模后,实验动物随机分为对照组、微重力模型组(生理盐水)、蛇床子素低浓度组(蛇床子素75 mg/kg)和蛇床子素高浓度组(150 mg/kg)。灌胃给药7天后,无菌条件下摘除脾脏,测量脾脏指数,流式细胞仪检测外周血Th1/Th2细胞比例,ELISA检测培养液TNF-α、IL-4、IL-5和IFN-γ含量,RT-PCR检测TNF-α、IL-4、IL-5和IFN-γ含量。结果蛇床子素组相较于微重力模型组,脾脏指数上升,外周血Th1/Th2比值明显升高(P<0.01)。TNF-α和IFN-γ细胞因子表达升高(P<0.01),而IL-4和IL-5表达下降(P<0.01)。结论蛇床子素能调节大鼠Th1细胞/Th2细胞的比例,具有免疫增强调节作用。 Objective To investigate the immunomodulatory effect of osthole on the balance of Th1 and Th2 cell under microgravity.Methods A microgravity model was established with 27 SD rats suspended from their hind limbs,and 9 SD rats were used as control.After 2 weeks,they were randomly divided into control group,microgravity control group(saline),low concentration group(osthole 75 mg/kg)and high concentration group(osthole 150 mg/kg).The spleen was aseptic removed and the peripheral blood was extracted.The proportion of Th1 and Th2 cells in the peripheral blood was measured by flow cytometry.The Th1/Th2 ratio was calculated.The levels of TNF-α,IL-4,IL-5,IFN-γwere detected by ELISA and RT-PCR.Results Compared with the microgravity group,the spleen index and the ratio of Th1 and Th2 in the peripheral blood of the osthole group increased(P<0.01).The expression of TNF-αand IFN-γcytokines increased(P<0.01),while the expression of IL-4 and IL-5 decreased(P<0.01).Conclusion Osthole has the function of immune enhancement and regulation,which can regulate the ratio of Th1/Th2 cell in rats.
