低氧环境下纤维生长因子2对人牙髓干细胞增殖及分化的影响

Effects of fiber growth factor 2 on proliferation and differentiation of human dental pulp stem cells in hypoxic environment

摘要 目的探究低氧环境下纤维生长因子2对人牙髓干细胞(human dental pulp stem cells,hDPSCs)增殖及分化的影响。方法收集hDPSCs并对其表面标记进行鉴定后,分别在常氧(21%O2)和低氧(3%O2)环境下,加或不加成纤维细胞生长因子2(fibroblast growth factor 2,FGF2)培养,MTT法分别检测各组细胞的增殖能力。对各组细胞进行成骨及成脂诱导培养21 d,观察低氧环境下纤维生长因子2对hDPSCs多向分化能力的影响。结果MTT结果显示,低氧+FGF2组hDPSCs在第2 d、3 d的增殖活性高于其余三组,低氧组及FGF2组次之,均高于常氧组;成骨诱导结果显示,低氧+FGF2组矿化结节最多,常氧组矿化结节最少;成脂诱导结果显示,低氧+FGF2组形成脂滴最多,常氧组形成脂滴最少。结论低氧环境下使用FGF2预处理培养能够提高hDPSCs的增殖能力表达,并可促进hDPSCs的分化。 Objective To investigate the effects of fibroblast growth factor 2(FGF2)on the proliferation and differentiation of human dental pulp stem cells(hDPSCs)in hypoxic environment.Methods hDPSCs were collected and identified the surface markers.Cells were cultured under normal oxygen(21%O2)and hypoxia(3%O2)with or without FGF2.The proliferation ability of cells in each group was detected by MTT method.After 21 days of osteogenesis and adipogenic induction,the effects of fibroblast growth factor 2 on the multidirectional differentiation of hDPSCs were observed under hypoxic conditions.Results MTT results showed that the proliferation activity of hDPSCs in hypoxia+FGF2 group was higher than that in the other three groups on the 2 nd and 3 rd day,followed by hypoxia group and FGF2 group,which were higher than that in normal oxygen group;osteogenesis induction results showed that mineralized nodules were the most in hypoxia+FGF2 group and the least in normal oxygen group;adipogenic induction results showed that lipid droplets were the most in hypoxia+FGF 2 group and the least in normal oxygen.Conclusion FGF22 pretreatment in hypoxic environment can improve the expression of hDPSCs and promote the differentiation of hDPSCs.
