芪冬颐心颗粒对心肌梗死大鼠心肌细胞凋亡及心脏微血管功能的影响 被引量:3

Effect of Qi Dong Yi Xin granule on myocardial cell apoptosis and cardiac microvascular function in myocardial infarction rats

摘要 目的研究芪冬颐心颗粒对心肌梗死大鼠心肌细胞凋亡及心脏微血管功能的影响。方法选用Wistar大鼠(250~300 g)160只,随机分为4组:假手术组、模型组、芪冬颐心颗粒组、地奥心血康组。模型组、芪冬颐心颗粒组、地奥心血康组通过结扎左冠状动脉主干制备心肌梗死模型,假手术组除不结扎冠状动脉外,其余操作与模型组完全相同。手术后第3天,芪冬颐心颗粒组按照150 mg/(kg·d)剂量灌胃给药,地奥心血康组按照150 mg/(kg·d)剂量灌胃给药,模型组和假手术组正常饲养。连续给药28天后通过超声心动图系统和BL-420F生物机能实验系统检测心功及血流动力学指标;HE染色法观察心肌细胞形态学变化;应用试剂盒检测心脏组织SOD、LDH活性,MDA、GSH、IL-18、IL-1β和NO的含量;通过Western blot检测Caspase-3、PECAM-1、eNOS和p-eNOS的蛋白表达。结果芪冬颐心颗粒组和地奥心血康显著提高了心肌梗死大鼠的心脏功能(P<0.01);减小心脏梗死面积;抑制氧化应激和炎症反应,进而抑制心肌细胞凋亡,同时,上调PECAM-1、p-eNOS蛋白表达(P<0.01)、NO含量,改善了心脏微血管功能。结论芪冬颐心颗粒通过抑制氧化应激损伤、炎症反应、改善心脏微血管功能发挥心肌保护作用。 Objective To observe the protective effect of Qi Dong Yi Xin granule on myocardial infarction rats.Methods One hundred and sixty Wistar rats(250~300 g)were used and randomly divided into 4 groups:sham group,model group,Qi Dong Yi Xin granule group and Di Ao Xin Xue Kang group.The left coronary artery trunk of model group was ligated to cause myocardial infarction,and the sham group had the same operations as the other groups except that the coronary artery was not ligated.Three days after the operation,the rats of Qi Dong Yi Xin granule group were intragastrically administered at 150 mg/(kg·d),and the rats of Di Ao Xin Xue Kang group were intragastrically administered at 150 mg/(kg·d),in addition,the rats of model group and sham group were fed normally.After 4 weeks of continuous administration,the cardiac function and hemodynamic parameters were measured by using the echocardiographic system and the BL-420 F biofunctional experiment system.Then,the heart was taken out and sliced.The morphology of myocardial cells were observed by HE staining.The activity of SOD,LDH and the content of MDA,GSH,IL-18,IL-1βand NO,were detected by using the kit.Western blot was used to detect the expression of PECAM-1,eNOS,p-eNOS and Caspase-3.Results Qi Dong Yi Xin granule group and Di Ao Xin Xue Kang significantly improved the cardiac function of myocardial infarction rats(P<0.01),reduced the area of heart infarction,inhibited oxidative stress and inflammatory response,thereby inhibited apoptosis of myocardial cells.At the same time,the expression of PECAM-1,p-eNOS protein and NO content were up-regulated(P<0.01),and the cardiac microvascular function was improved.Conclusion Qi Dong Yi Xin granules play a protective role in myocardium by inhibiting oxidative stress injury,inflammatory reaction and improving cardiac microvascular function.
