福建省龙岩市啮齿动物携带戊型肝炎病毒和温州病毒调查及基因特征

Infection status and genomic characterization of hepatitis E virus and Wenzhou virus in rodents in Longyan City,Fujian Province

目的调查福建省龙岩市啮齿动物戊型肝炎病毒(hepatitis E virus,HEV)和温州病毒(Wenzhou virus,WENV)感染情况及基因特征,为戊型肝炎病毒和温州病毒防控提供参考。方法在漳平市和武平县采集啮齿动物肠道组织样本,用巢式PCR扩增戊型肝炎病毒RdRp(RNA-dependent RNA polymerase)基因和温州病毒S(Small segment)基因。阳性PCR产物送交测序。采用生物信息学软件进行遗传特征分析。结果本调查共捕获141只啮齿动物。戊型肝炎病毒阳性率为7.8%,感染动物为黄胸鼠、黄毛鼠和针毛鼠,感染率分别为22.2%(2/9)、10.0%(8/80)和2.9%(1/35)。序列分析显示,龙岩市啮齿动物HEV与深圳啮齿动物及鼩鼱HEV关系较近,而与香港地区患者HEV分离株存在约14%的差异。所有啮齿动物均未检测出WENV。结论该研究是福建省首次在啮齿动物中发现HEV感染,对防控有重要意义;虽尚未在啮齿动物中发现WENV感染,但仍需进一步加强监测,以预防WENV感染流行。

Objective To investigate infection status and genomic characteristics of Hepatitis E virus(HEV)and Wenzhou virus(WENV)in rodents in Longyan City of Fujian Province,in order to provide reference for the prevention and control of HEV and WENV.Methods Rodent intestinal tissue samples were collected in Zhangping City and Wuping County.The RNA-dependent RNA polymerase(RdRp)gene of HEV and Small segment(S)gene of WENV were amplified by nested PCR.Positive PCR products were sent for sequencing.Genomic characterization was performed using bioinformatics software.Results A total of 141rodents were captured in the current study.The positive rate of HEV was 7.8%.The infected animals were R.losea,R.tanezumi,and N.fulvescens,with infection rates of 22.2%(2/9),10.0%(8/80)and 2.9%(1/35),respectively.Sequence analysis showed that the HEV found in rodents in Longyan City was closely related to HEV found in rodent and shrew from Shenzhen City,whereas there was about 14%difference compared to HEV isolates from Hong Kong region patients.WENV was not detected in any of the rodents.Conclusion This is the first study that detected HEV infection in rodents in Fujian Province,which has important implications for prevention and control.Although WENV infection was not detected in rodents in the current study,enhanced surveillance is still necessary to prevent potential WENV outbreaks.
