金属离子显色法对不同种属天然药物的抗氧化活性研究

摘要 目的建立了一种新型金属离子锰络合罗丹明B显色剂褪色反应体系(RB-Mn2+-H2O2)的分光光度法,用于检测不同种属的天然药物提取物对羟自由基(·OH)的抗氧化清除作用初步研究。方法以稀H2SO4作为溶剂介质,利用·OH迅速氧化RB使其化学褪色,在550nm处测定溶液吸光度变化,通过化学试剂褪色法对天然药物提取物进行了抗氧化性能实验。结果6种天然药物提取物对·OH具有明显的清除作用,清除率范围在27.58%~56.97%。结论该法简便灵敏,体系稳定,成本低廉,适于大批量天然药物抗氧化活性性能的筛选及其清除率的定量检测。 OBJECTIVE A new Fenton reaction system(RB-Mn2+-H2O2)spectrophotometric method was established for the determination of antioxidant scavenging effect of natural drug extracts on hydroxyl radicals by RB combined with manganese-hydrogen peroxide.METHODS RB was oxidized rapidly by·OH to make it fade chemically with H2SO4 as solvent medium.The absorbance of solution was measured at 550 nm.The antioxidant activity of natural medicines extracts was tested by chemical reagent fading method.RESULTS The extracts of 6 natural medicines had obvious scavenging effect on oOH,and the scavenging rate ranged from 27.58%to 56.97%.CONCLUSIONS The method is simple,sensitive,stable and low cost.It is suitable for screening antioxidant activity and quantitative detection of scavenging rate of large quantities of natural medicines.
