白藜芦醇后处理对脑缺血再灌注大鼠血脑屏障和ICAM-1、VCAM-1的影响 被引量:6

Effects of resveratrol post-treatment on the blood-brain barrier and ICAM-1、VCAM-1 in rats with cerebral ischemia-reperfusion

摘要 目的:探讨白藜芦醇后处理对脑缺血再灌注大鼠血脑屏障及ICAM-1、VCAM-1的影响。方法:随机将105只SD大鼠分为:假手术组,I/R组,Res组:低剂量组(10mg/kg/d)、中剂量组(20mg/kg/d)、高剂量组(40mg/kg/d)。线栓法制备大鼠MCAO模型,I/R后24h,观察大鼠神经功能评分,伊文思蓝法观察大鼠BBB通透性,干湿重法观察大鼠脑水肿,电镜观察大鼠BBB超微结构,ELISA法观察脑组织中ICAM-1、VCAM-1的表达。结果:高剂量组大鼠神经功能评分低于I/R组;I/R组脑组织中依文思蓝的含量高于假手术组,Res后处理组依文思蓝含量低于I/R组,Res高剂量组与I/R组比较有统计学意义;I/R组脑水肿程度高于假手术组,Res组脑水肿程度低于I/R组,Res高剂量组与I/R组比较有统计学意义;白藜芦醇组可以改善大鼠BBB超微结构,高剂量组更为显著;I/R组ICAM-1和VCAM-1的水平高于假手术组,Res组ICAM-1和VCAM-1的水平低于I/R组,Res高剂量组与I/R组比较有统计学意义。结论:白藜芦醇可能通过下调ICAM-1和VCAM-1,改善大鼠BBB通透性,改善BBB超微结构,减轻I/R后脑水肿程度,且具有剂量依赖性。 Objective To investigate the effects of post-treatment of resveratrol on BBB and the level of ICAM-1、VCAM-1 after cerebral I/R.Methods 105 SD rats were randomly divided into:sham group,I/R group,Res low-dose group,medium-dose group,high-dose group.MCAO model were prepared by the suture method.After 24h of I/R,the neurological function scores were observed.The permeability of BBB were observed by Evans blue method.Brain edema were observe by dry and wet weights,electron microscope to observe the ultrastructure of BBB,ELISA method to observe the expression of ICAM-1 and VCAM-1.Results The neurological scores in the high-dose group were lower than those in I/R group.The content of Evans blue in I/R group were higher than that in sham group,those in the Res groups were lower than that in the I/R group.Cerebral edema in I/R group were higher than that in sham group,those in Res group were lower than that in I/R group,high-dose were significant.Res groups can improve the ultrastructure of BBB,especially in the high-dose.The levels of ICAM-1 and VCAM-1 in I/R group were higher than those in sham group,the levels of ICAM-1 and VCAM-1 in Res groups were lower than those in I/R group,high-dose were significant.Conclusion Resveratrol may decrease the permeability of the BBB by down-regulating ICAM-1 and VCAM-1,improve the degree of ultrastructure of BBB,thereby reduce the degree of brain edema,and it is dose-dependent.
