侵袭性牙周炎与高龋患者唾液微生物对比 被引量:1

Comparison of Salivary Microorganism between Aggressive Periodontitis and High Caries Patients

摘要 目的:分析侵袭性牙周炎与高龋患者唾液微生物的差别。方法:选择我院2015年10月~2019年4月确诊并收治的侵袭性牙周炎患者17例及高龋患者15例作为牙周炎组及高龋组,收集各受试者唾液样本,采用十二烷基硫酸钠法提取样本中DNA,并以此进行扩增及变性梯度凝胶电泳,分析两组样本变性梯度凝胶电泳图谱特征、条带数量、戴斯相似性系数及微生物聚类分析结果。结果:侵袭性牙周炎组及高龋组分别有12个合格样本可用于后续研究;每位受试者唾液微生物变性梯度凝胶电泳图未出现完全相同的泳道,同一疾病组不同个体的唾液微生物变性梯度凝胶电泳图存在共有条带;侵袭性牙周炎组受试者变性梯度凝胶电泳图谱平均条带有(11.27±1.54)条,高龋组受试者变性梯度凝胶电泳图谱平均条带有(12.03±1.49)条,两组受试者变性梯度凝胶电泳图谱平均条带无明显差别;侵袭性牙周炎组与高龋组患者组间戴斯相似性系数明显低于侵袭性牙周炎组及高龋组患者各自组内戴斯相似性系数;侵袭性牙周炎组及高龋组受试者唾液微生物各自形成的聚类群具有明显区别。结论:侵袭性牙周炎患者与高龋患者唾液微生物群落构成具有明显差异,不同微生物的定植对口腔疾病具有重要影响。 Objective To investigate the difference of salivary microorganism between aggressive periodontitis and high caries patients.Methods 17 cases of aggressive periodontitis(AgP group)and 15 cases of high caries(HC group)treated in our hospital from October 2015 to April 2019 were selected.Non-irritating saliva samples were collected and DNA was extracted by sodium dodecyl sulfate(SDS).Then the denaturing gradient gel electrophoresis(DGGE)bands were cloned,sequenced and aligned.The DGGE characteristics,the number of bands,the Dyce similarity coefficient and the results of microbial community were analyzed.Results 12 qualified samples were collected in two groups.The DGGE had no exactly the same lane between groups,while the same group had the common bright band.The average band was(11.27±1.54)in the AgP group and(12.03±1.49)in the HC group,with no statistic difference.The similarity coefficient of two groups was lower than that of the single group.The salivary microbial community structure had significant difference between groups.Conclusion There is a significant difference in salivary microbial community structure between patients with caries or aggressive periodontitis,indicating that salivary microbial community structure maybe play a role in the inverse relationship between the two diseases.
