摘要
目的:探讨miR1251-3p负调控HE4对卵巢癌细胞株COC1的增殖和侵袭的影响。方法:实验分为空白组、阴性对照组、模拟物组(mimic)和抑制剂组(inhibitor)。采用qPCR、划痕实验和Transwell小室实验,检测miR-1251-3p在卵巢癌细胞株COC1细胞株侵袭转移中的作用。结果:miR-1251-3p与HE4的3’UTR区域有结合位点。与卵巢癌细胞株COC1空白组和阴性对照相比,转染miR-1251-3p mimics会抑制增殖和侵袭,而转染miR-1251-3p inhibitor可以促进卵巢癌细胞COC1的增殖和侵袭。结论:miR-1251-3p负调控HE4抑制卵巢癌细胞株COC1的增殖和侵袭。
Objective To investigate the effect of miR1251-3p negatively regulating HE4 on the proliferation and invasion of ovarian cancer cell line COC1.Methods The experiment was divided into blank group,negative control group,mimic group and inhibitor group.The role of miR-1251-3p in the invasion and metastasis of ovarian cancer cell line COC1 cell line was examined using qPCR,scratch test,and Transwell chamber experiment.Results miR-1251-3p had binding sites to the 3’UTR region of HE4.Compared with the ovarian cancer cell line COC1 blank group and negative control,transfection with miR-1251-3p mimics can inhibit proliferation and invasion,while transfection with miR-1251-3p inhibitor can promote proliferation and invasion of ovarian cancer cell COC1.Conclusion miR-1251-3p negatively regulates HE4 to inhibit the proliferation and invasion of ovarian cancer cell line COC1.
