摘要
目的:探讨雄鼠交配前高盐高脂高糖饮食(高复合饮食)两次暴露对子二代(F2)焦虑和抑郁样行为的影响。方法:亲代(F0)雄性Sprague-Dawley(SD)大鼠30只,随机分为对照饮食组(CD,n=15)和高复合饮食组(HD,n=15)。F0雄性大鼠交配前高复合饮食饲料喂养9周后与正常雌鼠交配产生子一代(F1),给予对照饮食喂养15周,从CD组和HD组的F1中随机取雄鼠交配前给予对照饮食或高复合饮食二次暴露9周,在第24周时F1雄鼠与正常的F1雌鼠交配产生F2,F2大鼠均给予普通饲料喂养。分别在第9、12、15、18、22和25周测量F2大鼠体重,在第12周和23周进行旷场实验、高架十字迷宫实验和新奇环境抑制摄食实验和在第25周进行强迫游泳实验。结果:F2大鼠体重检测结果显示雄鼠交配前高复合饮食两次暴露会导致F2大鼠体重增加,并以雌鼠体重增加更为明显。第23周旷场实验结果显示,高复合饮食两次暴露组(F0HD+F1HD)雌鼠和雄鼠的焦虑样行为比对照组(F0CD+F1CD)更明显。第12周和23周高架十字迷宫实验结果均显示,F0HD+F1HD组雌鼠焦虑样行为比F0CD+F1CD组更明显。第23周新奇环境抑制摄食实验结果显示,与F0CD+F1CD组相比,F0CD+F1HD组雌鼠和F0HD+F1CD组雄鼠的抑郁样行为更明显。第25周强迫游泳实验结果显示F0HD+F1HD组雌鼠的抑郁样行为显著高于F0CD+F1CD。结论:雄鼠交配前高复合饮食两次暴露会导致F2大鼠体重增加,以及产生焦虑和抑郁样行为,并且具有性别差异性,以雌鼠的体重增加和焦虑和抑郁样行为增加更为明显。
Objective To investigate the effects of feeding male rats before mating with high-salt-fat-sugar diet(high-complex diet)over two generations on anxiety and depression-like behavior in second generation offspring(F2).Methods Thirty male SD rats were randomly divided into a control diet group(CD,n=15)and a high-complex diet group(HD,n=15).F0 male rats were fed with a high-complex diet for 9 weeks before mating and mated with normal female rats to produce offspring(F1),given a control diet for 15 weeks,and then randomly selected male rats from F1 in the CD group and HD group,respectively.The control diet or the high-complex diet was exposed second time for 9 weeks before mating,and at the 24th week,F1 male rats mated with control diet feeding F1 female rats to produce F2 rats,all of which were fed with normal diet.F2 rats were measured for body weight at weeks 9th,12rd,15th,18th,22nd,and 25th.Open field test,elevated cross-maze test and novel environment inhibition feeding test at weeks 12nd and 23rd,and forced swimming test at week 25th were performed.Results The body weight test in F2 rats showed that male rats fed with high-complex diet over two generations before mating could lead to weight gain in F2 rats,and the weight gain of female rats was more significant.The results of open field test at week 23rd showed that F2 females and males of F0 and F1 male rats fed with high-complex diet group(F0HD+F1HD)was more significant than F2 rats of F0 and F1 male rats fed with control group(F0CD+F1CD).The results of elevated cross-maze test at weeks 12nd and 23rd showed that female rats in the F0HD+F1HD group had more anxiety-like behaviors than F0CD+F1CD group.The results of novel environment inhibition feeding test at week 23rdshowed that compared with F0CD+F1CD group,female rats in F0CD+F1HD group and male rats in F0HD+F1CD group had more depression-like behavior.The results of forced swimming test at week 25th showed that the depression-like behavior of female rats in F0HD+F1HD group was significantly higher than F0CD+F1CDgroup.Conclusion Male rats exposed to the high-complex diet twice before mating can lead to weight gain in F2 rats,as well as increase anxiety and depression-like behaviors,and with gender differention,with female rats gaining more weight and anxiety and depression-like behaviors.
