摘要
为获得花生栽培种中有效的InDel标记,本研究基于花生简化基因组检测到的InDel信息,经PCR扩增以及电泳检测验证引物的有效性与多态性,利用生物统计学软件检测InDel标记的多态性水平。结果表明,在花生基因组上设计的39对引物在覆盖4个植物学类型的21份花生中均可获得扩增产物,其中14对引物具有多态性,为总引物数的35.9%,有3对引物位于基因编码区;Shannon指数为0.1914~1.2951,均值为0.5794;多态性信息量(PIC)为0.0907~0.6746,均值为0.3496。本研究筛选获得的InDeI标记可为花生栽培种遗传图谱构建和分子标记辅助育种提供有效的遗传标记。
In order to screen the polymorphic InDel markers in peanut,PCR amplification and electrophoresis verification were carried following the reduced-representation genome sequencing(GBS).The result showed that total of 39 pair primers was successfully amplified.Based on 21 peanut cultivars of four typical botanical varieties 14 InDel markers showed the higher polymorphism and accounted for 35.9%of the total number of primers.Three of which were located in the coding regions.The Shannon′s index ranged from 0.1914 to 1.2951,with an average of 0.5794.The polymorphism information content(PIC)varied from 0.0907 to 0.6746,with an average of 0.3496.In summary,the InDel markers developed in this study could provide effective genetic marker for the construction of genetic map of cultivated peanut and molecular marker-assisted breeding in peanut.
