Expression Differences and Clinical Significance of FAM134B in Gastric Cancer Tissues

目的本研究分析134序列相似的家庭成员B(family with sequence similarity 134,member B,FAM134B)在胃癌组织中表达情况,并对胃癌组织中FAM134B的表达高低与胃癌患者的临床病理特征及总体生存率的关系进行分析。方法收集在作者医院进行手术并经组织学证明为胃癌组织及其配对的癌旁组织标本,对标本行蛋白免疫印迹(Western blot)试验明确FAM134B蛋白在胃癌组织和癌旁组织中的表达水平,利用Oncomine数据库荟萃分析FAM134B在胃癌中的表达情况;分别通过癌症基因组图谱(the cancer genome atlas,TCGA)数据库和Kaplan-Meier数据库来统计分析FAM134B表达水平的高低与胃癌患者临床病理特征和预后的关系。结果Western blot试验结果发现与癌旁组织相比,FAM134B蛋白在胃癌组织中的表达水平降低。通过荟萃分析Oncomine数据库中的5项研究,共包括478个样本,发现FAM134B基因在胃癌中的表达水平降低,差异具有统计学意义(P=0.045)。通过TCGA数据库分析440例胃癌患者临床病理特征发现,FAM134B的表达量与病理分期相关,差异具有统计学意义(P<0.05),与年龄、性别、T分期、N分期、M分期无明显相关性(P>0.05)。运用Kaplan-Meier数据库分析875例胃癌患者的生存情况发现,FAM134B表达水平与胃癌患者总体生存率相关(P<0.05),FAM134B低水平表达组胃癌患者生存时间更短,预后更差。结论FAM134B在胃癌组织中表达水平下降,且FAM134B低水平表达的胃癌患者病理分期更晚、预后更差。总之FAM134B在胃癌组织中一定程度上表现出抑癌基因的特性。

Objective To analyze the expression of family with sequence similarity 134,member B(FAM134 B)in gastric cancer tissues,and analyze the relationship among the high and low expression of FAM134 B in gastric cancer tissues and the clinicopathologic features and overall survival rate of patients with gastric cancer.Methods The samples of gastric cancer tissues and their paired adjacent tissues that were histologically proven to be gastric cancer and underwent surgery in author′s hospital were collected.The expression level of FAM134 B protein in gastric cancer tissues and adjacent tissues was determined by Western blot.The expression of FAM134 B gene in gastric cancer was analyzed by Oncomine database,the relationship between the high and low expression level of FAM134 B and the clinicopathologic features and prognosis of patients with gastric cancer was statistically analyzed by the cancer genome atlas(TCGA)database and Kaplan-Meier database.Results Western blot showed that the expression level of FAM134 B protein in gastric cancer was lower than that in adjacent tissue.Five studies in Oncomine database included 478 samples were analyzed by Meta-analysis.It was found that the expression level of FAM134 B gene in gastric cancer decreased,and the difference was statistically significant(P=0.045).It could be found that the expression of FAM134 B was related to the pathological stage,and the difference is statistically significant(P<0.05)by analyzing the clinicopathologic features of 440 gastric cancer patients in TCGA database,and there was no significant correlation with age,gender,T stage,N stage and M stage(P>0.05).The survival situation of 875 patients with gastric cancer was analyzed by Kaplan-Meier database.It was found that the expression level of FAM134 B was correlated with the overall survival rate of gastric cancer patients,and the difference was statistically significant(P<0.05).The survival time of gastric cancer patients in the low level expression group of FAM134 B was shorter and the prognosis was worse.Conclusion The expression level of FAM134 B in gastric cancer tissues decreased,and the gastric cancer patients with low expression of FAM134 B has later pathological stage and worse prognosis.In conclusion,FAM134 B shows the characteristics of tumor suppressor gene in gastric cancer tissues to a certain extent.
