基于生物信息学分析SLC44A4基因在乳腺癌中的表达及其临床意义

Expression and Clinical Value of SLC44A4 Gene in Breast Cancer Based on Bioinformatics Analysis

目的通过分析生物信息数据库,探讨胆碱转运蛋白样蛋白SLC44A4基因在乳腺癌中的表达及其临床意义。方法检索Oncomine数据库,分析SLC44A4基因在不同类型肿瘤中的表达情况;检索基因表达谱动态分析(gene expression profiling interactive analysis,GEPIA)数据库,对比乳腺癌组织与正常乳腺组织之间SLC44A4基因的表达差异,分析该基因的表达水平与乳腺癌临床病理分期的关系。利用Kaplan-Meier Plotter在线分析SLC44A4基因表达水平与乳腺癌患者预后的关系。检索肿瘤单细胞(CancerSEA)数据库分析SLC44A4基因与乳腺癌单个细胞功能状态的相关性。结果SLC44A4在多种肿瘤组织中高表达。与正常乳腺组织相比,乳腺癌组织中SLC44A4基因的表达明显升高(P<0.05),且不同临床病理分期和分型的乳腺癌患者SLC44A4基因表达水平比较差异有统计学意义(P<0.05)。从总体来看,SLC44A4基因高表达组总生存(overall survival,OS)期高于低表达组(P<0.05)。此外,SLC44A4基因的表达与乳腺癌细胞的侵袭、细胞周期、DNA修复呈正相关性。结论SLC44A4基因在乳腺癌组织中高表达,与患者预后相关,并与乳腺癌细胞功能存在一定相关性。

Objective To analyze the expression of choline transporter-like protein SLC44A4 gene in breast cancer and its clinical value based on bioinformatics database analysis.Methods The Oncomine database was searched to analyze the expression of SLC44A4 gene in different types of tumors.The gene expression profiling interactive analysis(GEPIA)database was used to compare the difference of SLC44A4 gene expression between breast cancer and normal tissues,and to analyze the relationship between the expression level of SLC44A4 gene and the clinicopathological stage of breast cancer.Kaplan-Meier Plotter was used to analyze the relationship between the expression level of SLC44A4 gene and the prognosis of breast cancer patients online.The correlation between SLC44A4 gene and functional states of single breast cancer cells was analyzed using CancerSEA database.Results SLC44A4 was highly expressed in many kinds of tumor tissues.The expression level of SLC44A4 gene in breast cancer tissues was significantly higher than that in normal tissues(P<0.05).There was significantly different in the expression of SLC44A4 gene among breast cancer patients with different clinicopathological stages and types(P<0.05).The overall survival(OS)of the SLC44A4 gene high expression group was higher than that of the low expression group(P<0.05).In addition,SLC44A4 gene expression was positively correlated with invasion,cell cycle and DNA repair of breast cancer cells.Conclusion SLC44A4 gene is highly expressed in breast cancer tissue,which is related to the prognosis of patients and has a certain correlation with the function of breast cancer cells.
