舰船非金属材料燃烧释放的烟雾对小鼠肺组织的影响

摘要 目的观察密闭舱室内非金属材料燃烧释放的烟雾对吸入后小鼠肺组织的损伤效应。方法成年雄性ICR小鼠30只,体质量(22±2)g。将小鼠随机分为3组,即0.5 h烟雾染毒组、24 h烟雾染毒组和对照组,每组各10只。以HOPE-MED 8050型动态染毒系统为依托,建立非金属材料燃烧释放烟雾经呼吸道吸入暴露小鼠模型。酶联免疫吸附试验(enzyme linked immunosorbent assay,ELISA)检测肺组织匀浆肿瘤坏死因子α(tumor necrosis factor alpha,TNF-α)、白细胞介素1(interleukine 1,IL-1)、IL-6、IL-8、IL-10炎性因子水平。化学发光比色法检测肺组织内超氧化物歧化酶(superoxidedismutase,SOD)、一氧化氮合酶(nitric oxide synthase,NOS)、谷胱甘肽(glutathione,GSH)、髓过氧化物酶(myeloperoxidase,MPO)活性,及丙二醛(malondialdehyde,MDA)含量。结果与对照组比较,0.5 h和24 h烟雾染毒组小鼠肺组织内SOD和GSH活性明显降低(P<0.01),而NOS及MPO的活性明显升高(P<0.01),MDA含量明显增高(P<0.01)。肺组织内炎症因子TNF-α、IL-8、IL-6和IL-1的含量显著升高(P<0.01),IL-10的含量显著下降(P<0.01)。与0.5 h烟雾染毒组比较,24 h烟雾染毒组小鼠肺组织中SOD和GSH活性明显降低(P<0.01),NOS及MPO的活性明显升高(P<0.05),MDA含量明显增高(P<0.01),炎症因子含量更高(P<0.01),IL-10的含量显著下降(P<0.05)。结论非金属材料燃烧释放的烟雾可导致小鼠吸入性肺损伤,且染毒后24 h损伤更严重。 Objective To observe the inhalation injury effects of smoke released by non-metallic materials burning on the lung tissue of mice.Methods Thirty adult male ICR mice,weighing(22±2)g.The mice were randomly divided into 3 groups,namely 0.5 h smoke-exposed group,24 h smoke-exposed group and control group,each with 10 mice.Based on HOP-MED 8050 dynamic poisoning system,a mouse model of smoke emission from non-metallic materials was established by inhalation exposure through respiratory tract.The levels of inflammatory factors of lung tissue homogenate tumor necrosis factor alpha(TNF-α),interleukine 1(IL-1),IL-6,IL-8,IL-10 were detected by enzyme linked immunosorbent assay(ELISA)method.Chemiluminescence colorimetry to detect the activity of superoxide dismutase(SOD),nitric oxide synthase(NOS),glutathione(GSH),myeloperoxidase(MPO)in lung tissue,and malondialdehyde(MDA)content.Results Compared with control group,the activities of SOD and GSH in the lung tissue of 0.5 h and 24 h smoke-exposed group were significantly reduced(P<0.01),while the activities of MPO and NOS were significantly increased(P<0.01),and the MDA content was significantly increased(P<0.01).The content of inflammatory factors TNF-α,IL-8,IL-6 and IL-1 in lung tissue increased significantly(P<0.01),and the content of IL-10 decreased significantly(P<0.01).Compared with 0.5 h smoke-exposed group,the activities of SOD and GSH in lung tissue homogenate of mice in the 24 h smoke-exposed group were significantly decreased(P<0.01),the activities of MPO and NOS were significantly increased(P<0.05),and the MDA content was significantly increased(P<0.01),inflammatory factors significantly increased(P<0.01),and the content of IL-10 significantly decreased(P<0.05).Conclusion The exposure in smoke released by non-metallic materials burning can cause aspiration lung injury in mice,and the injury is more serious 24 hours after exposure.
