多梳家族基因PCGF1沉默对胶质母细胞瘤的调控作用及其生存预后分析

Regulation of Glioblastoma by Silencing of Polycomb Group Gene PCGF1 and Analysis of Its Survival Prognosis

摘要 目的探讨多梳家族因子1(polycomb group factor 1,PCGF1)表达与胶质母细胞瘤(glioblastoma,GBM)恶性程度及生存预后的关系。方法采用生物信息学软件分析评价GBM中PCGF1 mRNA表达水平及其与GBM患者生存预后关系;通过免疫组织化学方法检测65例样本组织中PCGF1的表达。抑制PCGF1表达,检测细胞增殖能力和周期变化。结果PCGF1 mRNA表达水平在GBM组织中显著上调,并且与患者生存预后密切相关;进一步免疫组化实验证实,与低度恶性GBM相比,高度恶性GBM中PCGF1的表达上调更为明显。PCGF1沉默,细胞增殖能力下降,G1期细胞比例明显增加。结论PCGF1的表达与GBM恶性程度及生存预后密切相关,PCGF1可能是GBM诊断、生存及预后的潜在生物标志分子。 Objective To investigate the relationship between the expression of polycomb group factor 1(PCGF1)and glioblastoma(GBM)malignancy and survival prognosis.Methods Bioinformatics software was used to analyze and evaluate the PCGF1 mRNA expression level in GBM and its relationship with the survival and prognosis of GBM patients.The expressions of PCGF1 in 65 samples of tissues were detected by immunohistochemistry.The cell proliferation and cell cycle changes were detected after inhibiting PCGF1 expression.Results The expression level of PCGF1 mRNA was significantly up-regulated in GBM tissues,and was closely related to the survival prognosis of patients.Furtherly,immunohistochemical experiments confirmed that compared with low-grade GBM,PCGF1 expression was up-regulated more significantly in high-grade GBM.The proliferation ability of PCGF1 silenced cells decreased,and the proportion of cells in G1 phase increased significantly.Conclusion The expression of PCGF1 is closely related to the malignancy and survival prognosis of GBM.PCGF1 may be a potential biomarker for the diagnosis,survival and prognosis of GBM.
