白藜芦醇对PM2.5致大鼠肺损伤的抑制作用 被引量:3

摘要 目的探讨白藜芦醇对PM2.5染毒致大鼠肺损伤的抑制作用。方法 64只SPF级SD大鼠(雌、雄各半)随机分成溶剂对照组(吐温80)、白藜芦醇对照组(40 mg/kg)及低、中、高剂量PM2.5组(分别为2、4、8 mg/kg)和白藜芦醇+低、中、高剂量PM2.5组(白藜芦醇灌胃28 d+气管滴注相应剂量PM2.5),共8组,每组8只,其中PM2.5染毒为两天1次,共4次。末次染毒24 h后,以10%水合氯醛腹腔注射麻醉,测定大鼠肺组织中谷胱甘肽(GSH)、丙二醛(MDA)含量和谷胱甘肽过氧化物酶(GSH-Px)、总超氧化物歧化酶(T-SOD)活力,测定股动脉血中白细胞、中性粒细胞和淋巴细胞水平,并观察各组大鼠肺组织的病理学变化。结果实验进行第30、32、34天,各剂量PM2.5组和白藜芦醇+中、高剂量PM2.5组大鼠体温高于溶剂对照组,白藜芦醇+中、高剂量PM2.5组大鼠体温高于白藜芦醇对照组,白藜芦醇+各剂量PM2.5组体温低于相应剂量PM2.5组,差异均有统计学意义(P<0.05)。与溶剂对照组比较,各剂量PM2.5组大鼠血液中白细胞、中性粒细胞和淋巴细胞升高,肺组织中GSH含量、GSH-Px及T-SOD活力下降,MDA含量升高,差异均有统计学意义(P<0.05);与相应剂量PM2.5组比较,白藜芦醇对照组和白藜芦醇+各剂量PM2.5组大鼠血液中白细胞、中性粒细胞和淋巴细胞下降,肺组织中GSH含量、GSH-Px及T-SOD活力升高,MDA含量下降,差异均有统计学意义(P<0.05)。结论 PM2.5可引起大鼠肺组织出现病理性损伤,导致机体体温、血象及氧化应激等指标发生改变,而白藜芦醇对PM2.5引起的肺损伤有明显的抑制作用。 Objective To understand the inhibitive effects of resveratrol on PM2.5-induced lung damage in rats. Methods A total of 64 adult SD rats of SPF grade,half male and half female,were randomly divided into eight groups,eight in each,solvent control group(polysorbate-80),resveratrol group(40 mg/kg),PM2.5 exposure groups(2 mg/kg,4 mg/kg and 8 mg/kg),resveratrol +PM2.5 groups(the low,middle and high experimental dose). Resveratrol was given through gavage for 28 days,followed by PM2.5 exposure conducted by intratracheal instillation,once two days,a total of four times. Twenty-four hours after the last treatment,10% chloral hydrate was used for anaesthetization. The contents of GSH and MDA,the activities of GSH-Px and T-SOD were determined,the number of leukocyte,percentage of neutrophils and lymphocytes in arterial blood were observed,the pathological examination on lung tissue was also performed. Results At the 30 th,32 nd,34 th day of the treatment,higher body temperatures were found in PM2.5 exposure groups,resveratrol +moderate and high dose of PM2.5 groups compared with solvent control group,as well as in resveratrol+moderate and high dose of PM2.5 groups compared with resveratrol control group,also in resveratrol +PM2.5 exposure groups compared with PM2.5 exposure groups with the same dose(P <0.05).More leukocytes,higher percentage of neutrophils and lymphocytes in blood,lower contents of GSH,higher activities of GSHPx and T-SOD,higher content of MDA in lung tissue were found in PM2.5 exposure groups compared with those in solvent control group(P<0.05). Less leukocytes,higher percentage of neutrophils and lymphocytes in blood,higher contents of GSH,higher activities of GSH-Px and T-SOD,lower content of MDA in lung tissue were found in resveratrol+PM2.5 exposure groups compared with those in groups with the same exposure doses of PM2.5(P<0.05). Conclusion PM2.5 exposure can cause the lung damage,and can cause the variation of body temperature,blood picture and oxidative stress in rats,which can be inhibited by resveratrol.
