邻苯二甲酸二甲酯对小鼠肝脏Ca^2+-Mg^2+-ATP酶和Na^+-K^+-ATP酶的影响 被引量:4

Effects of dimethyl phthalate exposure on Ca^2+-Mg^2+-ATPase and Na^+-K^+-ATPase activity in liver of mice

摘要 目的探讨邻苯二甲酸二甲酯(DMP)对小鼠肝脏Ca2+-Mg2+-ATP酶和Na+-K+-ATP酶活力的影响。方法将80只雌雄各半的健康清洁级C57BL/6J小鼠随机分为对照(纯玉米油)组和0.5、1.0、2.0 g/kg DMP染毒组,每组20只,雌雄各半。采用灌胃的方式进行染毒,灌胃体积为10 ml/kg,每天灌胃1次。分别在灌胃20、40 d后,每组选择10只小鼠(雌雄各半)测定肝脏Ca2+-Mg2+-ATP酶和Na+-K+-ATP酶的活力。结果灌胃20 d时,与对照组比较,各剂量DMP染毒组雄性小鼠肝脏的Ca2+-Mg2+-ATP酶活力以及1.0、2.0 g/kg DMP染毒组雄性小鼠肝脏的Na+-K+-ATP酶活力均较低,差异有统计学意义(P<0.05);0.5、2.0 g/kg DMP染毒组雌性小鼠肝脏的Ca2+-Mg2+-ATP酶活力以及2.0 g/kg DMP染毒组雌性小鼠肝脏的Na+-K+-ATP酶活力均较高,差异有统计学意义(P<0.05)。灌胃40 d时,与对照组比较,各剂量DMP染毒组雄性小鼠肝脏Ca2+-Mg2+-ATP酶活力无明显变化,0.5 g/kg DMP染毒组雄性小鼠肝脏的Na+-K+-ATP酶活力较高,差异有统计学意义(P<0.05);各剂量DMP染毒组雌性小鼠肝脏的Ca2+-Mg2+-ATP酶和Na+-K+-ATP酶活力均较低,差异有统计学意义(P<0.05)。结论DMP经口染毒对小鼠肝脏的Ca2+-Mg2+-ATP酶和Na+-K+-ATP酶活力具有干扰作用,雌性小鼠比雄性小鼠更敏感。 Objective To understand the effects of dimethyl phthalate(DMP)on Ca2+-Mg2+-ATPase and Na+-K+-ATPase activity in liver of mice.Methods Eighty C57 BL/6 J mice,half female and half male,were randomly divided into four groups,20 in each,among which the exposure groups were treated with DMP at the doses of 0.5,1.0 and 2.0 g/kg respectively and the control group was treated with pure corn oil,through gavage at 10 ml/kg,once a day.After 20 days and 40 days of treatment,10 male and female mice in each group were collected respectively,the liver tissues were collected for the measurement of the total protein content,the activities of Ca2+-Mg2+-ATPase and Na+-K+-ATPase in the homogenate.Results After 20 days of DMP exposure,the Ca2+-Mg2+-ATPase activity in male mice of each dose group was lower than that in the control group(P<0.05),the Na+-K+-ATPase activities in the 2.0 g/kg DMP and 1.0 g/kg DMP dose groups was significantly lower than those in the control group(P<0.05);Ca2+-Mg2+-ATPase activities in female mice of 2.0 g/kg DMP and 0.5 g/kg DMP dose groups were higher than those in the control group(P<0.05),and Na+-K+-ATPase activity in the 2.0 g/kg DMP dose group was significantly higher than that in the control group(P<0.05).After 40 days of DMP exposure,there was no significant difference in the Ca2+-Mg2+-ATPase activity between the male mice(P>0.05),and the Na+-K+-ATPase activity in the 0.5 g/kg DMP dose group was higher than that in the control group(P<0.05);The activities of Ca2+-Mg2+-ATPase and Na+-K+-ATPase in female mice were significantly lower than those in the control group(P<0.05).Conclusion DMP can interfere with the Ca2+-Mg2+-ATPase and Na+-K+-ATPase activities in the liver of mice,and female mice are more sensitive than male mice.
