The gender difference of myosin in bladder detrusor

目的:探讨不同性别之间膀胱逼尿肌肌球蛋白的表达差异。方法:人正常膀胱逼尿肌和男性前列腺标本来自于武汉大学中南医院膀胱癌全切患者以及心脏死亡患者,病理证实为正常膀胱逼尿肌或前列腺组织的标本,而正常腹主动脉、下腔静脉和阴茎海绵体(CC)组织均为供体捐献;根据性别将正常膀胱逼尿肌标本分为两组;竞争性RT‐PCR检测SMM同工异构体(SM‐A/‐B,SM1/2和肌球蛋白轻链MLC17a/b)的表达;实时定量RT‐PCR检测膀胱SMM重链(MHC)和NMM重链的三种同工异构体(NA,NB和NC)的表达。结果:共收集35例人膀胱逼尿肌标本,5例腹主动脉、5例下腔静脉、5例阴茎海绵体(CC)及5例男性前列腺组织;我们发现膀胱逼尿肌SM表达约20%SM‐B,45%SM2和70%MLC17a;腹主动脉表达约3%SM‐B,50%SM2和26%MLC17a,下腔静脉表达约5%SM‐B,51%SM2和39%MLC17a,CC表达约10%SM‐B,51%SM2和43%MLC17a,男性前列腺组织表达约22%SM‐B,47%SM2和89%MLC17a;另外,女性膀胱逼尿肌表达较多的SM‐B及MLC17a,较少的NA和NC。结论:女性膀胱逼尿肌表达较多的SM‐B及MLC17a,较少的NA和NC,可能与女性膀胱过度活动症高发病率相关。

Objective:To explore the different expression of myosin in bladder detrusor between genders.Methods:Human bladder dutrusor and prostate tissues were collected from the patients treated with radical resection of bladder cancer and from donors of heart death in Zhongnan Hospital of Wuhan University.Meanwhile,the normal abdominal aorta,vein and corpus cavernosum(CC)tissues were collected from donors of heart death.SMM isoforms,including SM myosin heavy chain(MHC)isoforms(SM1/2 and SM-A/B)and myosin light chain 17 isoforms(LC17a/b),were determined via competitive RT-PCR.MHC and NMM isoforms(NA,NB,and NC)were further analyzed via real-time RT-PCR.Results:A total of 35 bladder detrusor tissues were collected with 5 abdominal aorta,5 abdominal vein,5 corpus cavernosum(CC),and 5 prostate specimens collected.At the mRNA level,bladder dutrusor expressed approximately 20%SM-B,70%MLC17a,and 45%SM2.Aorta and vena cava expressed approximately 3%SM-B,50%SM2,26%MLC17a,5%SM-B,51%SM2,and39%MLC17a,respectively.Meanwhile,CC expressed approximately 10%SM-B,51%SM2 and43%MLC17a,and male prostate tissues expressed approximately 22%SM-B,47%SM2,and 89%MLC17a.In addition,female bladder detrusor expressed higher SM-B and MLC17awith less expressions of NA and NC.Conclusion:SM-B and MLC17aare more expressed in female detrusor with the less expressions of NA and NC,which might be related to the high incidence of overactive bladder in females.
