间质成纤维细胞激活蛋白对胃癌细胞SGC7901生物学行为的作用 被引量:1

Effect of stromal fibroblast activation protein alpha on the biological behaviors of gastric cancer cell SGC7901

摘要 目的:研究间质微环境内成纤维细胞激活蛋白(FAP)对胃癌细胞SGC7901增殖、迁移、侵袭和抗凋亡能力的作用。方法:体外实验中,建立过表达FAP的人胚肺成纤维细胞(HELFFAP)和阴性对照(HELFNC),分别与SGC7901细胞共培养(FAP和NC组),通过CCK8、Transwell侵袭/转移以及流式细胞学实验研究间质FAP对其增殖、迁移、侵袭和凋亡的影响。结果:共培养72 h后,HELFFAP细胞促进SGC7901细胞增殖、迁移、侵袭,并抑制其细胞凋亡。结论:间质微环境来源的FAP能够促进胃癌细胞的增殖、迁移和侵袭能力,抑制细胞凋亡。 Objective:To study the effect of stromal fibroblast activated protein alpha(FAP)on the proliferation,migration,invasion,and apoptosis of gastric cancer cell SGC7901.Methods:Human embryonic lung fibroblasts(HELF)cells were transfected with lenti-virus overexpressed of FAP(FAP group)and negative control(NC group)in vitro,then they were co-cultured with SGC7901 cells for 72 h.The effects of stromal FAP on the proliferation,migration,invasion and apoptosis of SGC7901 cells were detected by CCK8 assay,Transwell migration,invasion assay,and flow cytometry,respectively.Results:Coculture with HELFFAP cells for 72 h,the proliferation,migration,invasion,and antiapoptosis abilities of SGC7901 cell were significantly promoted.Conclusion:Stromal FAP could promote the proliferation,migration,invasion as well as the anti-apoptosis abilities of gastric cancer cells in vitro.
