优质野生宽叶黑果枸杞种质高效离体快繁体系建立及优化研究

Establishment and optimization of propagation system for wild Lycium ruthenicum germplasm in vitro with high-efficiency and high-quality

摘要 为建立优质野生宽叶黑果枸杞离体培养与快速繁殖技术新体系,该文以野生宽叶黑果枸杞的带芽茎段为外植体,筛选优化适宜的芽诱导培养基,芽增殖协同生根的一步成苗培养基,优化炼苗、移栽等配套技术体系。结果显示:以MS+1.5mg/L6-BA+0.3mg/L NAA+50mg/L ASA+10g/L PVA为芽诱导最佳培养基;以MS+1.0mg/L 6-BA+0.8mg/L NAA+150mg/L ASA+20g/L PVA为芽最佳增殖和生根培养基,可一次成苗,且成苗时间短、苗齐苗壮,成活率达到97.6%。 The aim of this study is to establish a new technology system in which wild Lycium ruthenicum could be cultured and propagated rapidly in vitro with high-quality.Using the bud-bearing stems of wild Lycium ruthenicum as explants,two types of mediums were selected and optimized,one was used for bud induction and the other for one-step seedling in which bud multiplication was accompanied rooting.Then the technical systems for seedling refinement and transplanting was optimized.The best medium for bud induction is MS+1.5 mg/L 6-BA+0.3 mg/L NAA+50 mg/L ASA+10 g/L PVA.The best medium for bud multiplication and rooting is MS+1.0 mg/L 6-BA+0.8 mg/L NAA+150 mg/L ASA+20 g/L PVA.When the two optimal medium are used in combination,seedlings could grow at one time,meanwhile the seedling time is short,the seedlings are full and strong with 97.6%survival rate.A new technology system for propagation of wild Lycium ruthenicum in vitro with high-efficiency and high-quality was established,which could provide technical support for the large-scale propagation and rapid promotion of superior germplasm of Lycium ruthenicum.
