摘要
以适宜重庆地区广泛栽培的隋珠草莓优良品种为对象,以隋珠草莓的茎尖为外植体,优化了不同植物激素组合对不定芽诱导、增殖及生根的影响,同时也优化了移栽基质配方对组培苗驯化称移栽成活的影响。结果显示,MS+6-BA 1.0 mg/L+IBA 0.1 mg/L+蔗糖30 g/L为茎尖不定芽诱导的最佳培养基,MS+6-BA 1.0 mg/L+IBA 0.1 mg/L+蔗糖30 g/L为不定芽增殖壮苗的最佳培养基,1/2MS+IBA 0.2 mg/L+蔗糖30 g/L为不定芽生根的最佳培养基,草炭土∶泥土∶珍珠岩=5∶4∶1为草莓组培苗移栽成活率最高的移栽基质。本研究为隋珠草莓优良品种工厂化快速繁育种苗提供了技术支撑。
‘Suizhu’is a wildly grown strawberry species in Chongqing.In order to establish the optimal tissue culture and rapid propagation techniques for‘Suizhu’,the shoot of which was selected as explants,and the effects of plant growth regulators on bud induction,proliferation,as well as rootage were evaluated.In addition,the effect of cultivation medium on survival ratio of potting was also estimated.The results showed that,the suitable medium for bud induction is MS+1.0 mg/L 6-BA+0.1 mg/L IBA+30 g/L sucrose;The optimal media for bud proliferation is MS+1.0 mg/L 6-BA+0.1 mg/L IBA+30 g/L sucrose;Furthermore,application of 1/2 MS+0.2 mg/L IBA+30 g/L sucrose shows the optimal effect on rootage;The highest survival ratio of potting results from the cultivation medium(peat soil∶soil∶perlite=5∶4∶1).Overall,the results serve as an excellent base for large-scale breeding of‘Suizhu’strawberry through the tissue culture and rapid propagation techniques.
