摘要
比较不同公司甘露聚糖酶(A、B和C)的酶学性质,为甘露聚糖酶在各个领域的应用提供理论基础。通过对3种甘露聚糖酶进行耐受性、耐温性等处理,采用DNS比色法检测酶活存留率对3种酶进行理化性质评估。酶A、B和C的最适pH相同,均为2.50;酶B和C的最适温度相同为70℃,而酶A为75℃;热稳定性酶C最好,85℃处理后仍保持40%以上的酶活存留率;酶C对胃-胰蛋白酶的耐受效果最好,对pH的耐受性酶B最好,在pH 2.50~11.50的的范围内酶活存留率高于60%,此外,3种甘露聚糖酶均具有较强的耐酸性。3种甘露聚糖酶可根据不同行业对甘露聚糖酶的温度、pH要求,应用于环境、食品、医药等领域中。
To compare the enzymatic properties of mannanase from three different companies to provide a theoretical basis for mannanase applications in various social fields.The three of mannanases were treated with tolerance and temperature tolerance;the physicochemical properties of the three enzymes were evaluated by DNS colorimetry.The optimum pH of enzymes A,B and C is the same as 2.50;the optimum temperature of enzyme B and C was 70℃,while enzyme A was 75℃;the enzyme C exhibited the best thermal stability,more than 40%survival rate of enzyme activity after treatment at 85℃;Enzyme C showed the best tolerance to gastric-trypsin,enzyme B showed the best tolerance to pH and the survival rate of enzyme activity was higher than 60%in the broad range of pH 2.50~11.50.In addition,all three mannanases showed strong acid-resistance.Three kinds of mannanase can be used in environment,food,medicine and other fields according to the temperature and pH requirements of different industries.
