摘要
目的初步研究促卵泡激素(果纳芬和普丽康)对胆固醇代谢的影响。方法建立肝S9体外孵育体系,向体系中加入浓度为0.025,0.25,2.5 IU/ml的两种重组促卵泡激素注射液,并运用UPLC-MS/MS测定胆固醇浓度。结果与空白组相比,两种FSH在加入浓度为0.25 IU/ml重组促卵泡激素后,胆固醇的浓度与空白组胆固醇浓度的差值随着作用时间的增加而增大,当加入2.5 IU/ml的重组促卵泡激素后,胆固醇的浓度与空白组胆固醇浓度的差值随着作用时间的增加呈先增加后减小的趋势。(0~40 min呈增加趋势,40~80 min呈减小趋势)。结论在该试验条件下,促卵泡激素能够促进胆固醇代谢,且低浓度促卵泡激素(0.25IU/ml)较高浓度促卵泡激素(2.5 IU/ml)促进胆固醇代谢作用更为显著。
Objective To explore the effect of follicle-stimulating hormone(Gonal-F and Puregon)on cholesterol metabolism.Methords The rat liver S9 incubation system were conducted.Different concentrations of recombinant follitropin for injection 0.025,0.25,2.5 IU/ml were added in the incubations.Cholesterol was extracted by protein precipitation and analyzed by UPLC-MS/MS.Trend of peak area over time was utilized to monitor change of cholesterol metabolism.Results Compared with the blank group,peak area was smaller after recombinant follitropin injection added in the incubations.Difference in peak area of cholesterol between the 0.25 IU/ml group and the blank group showed an increase in 0~80 min,while that between the 2.5 IU/ml group and the blank group showed an increase in 0min^40min but a decrease in 40~80 min.Conclusions Under this experimental condition,follicle-stimulating hormone could promote the cholesterol metabolism.Low concentration of follicle-stimulating hormone(0.25 IU/ml)played a stronger role in promoting cholesterol metabolism than the high concentration(2.5IU/ml).
